Sangamo BioSciences, Inc. (Nasdaq: SGMO) announced the presentation of Phase 2 clinical data from its ZFP Therapeutic program to develop SB-509, a zinc finger protein transcriptional activator (ZFP-TF) of the vascular endothelial growth factor (VEGF)-A gene as a treatment for ALS. The data from study SB-509-801 demonstrated that the drug was well-tolerated in subjects with ALS and that 40% of SB-509 treated subjects had delayed deterioration of toe and ankle muscle strength as measured by manual muscle testing (MMT) compared to 23% of baseline-matched historic controls. In addition, positive improvements in electrophysiological measures of motor nerves were observed in a subset of treated subjects. The data were presented at the Society for Neuroscience Annual Meeting which is being held in San Diego.

"These data are consistent with the delay in muscle deterioration that we have observed in both preclinical models of ALS as well as in clinical studies of this drug in subjects with diabetic neuropathy, a condition that is also characterized by degeneration and deterioration of nerve function," stated Dale Ando, M.D., Sangamo's vice president of therapeutic development and chief medical officer. "Our primary aim in this trial was to assess safety and tolerability of treatment with SB-509 in this vulnerable patient population and to conduct a signal-seeking study in which we assessed the effect of treatment on several clinical measures of function in ALS. As this was an early-stage trial, for safety reasons we treated subjects twice over a 90 day period, although our positive preclinical animal data were achieved with a more frequent dosing schedule. With that said, the delayed deterioration in muscle strength in the ankle and toe are very encouraging and may reflect the importance of focused regional dosing, as the leg muscles received the highest volume of SB-509 compared to the neck and arms. These data provide valuable information for the design of future studies which would include more frequent and focused dosing with SB-509."

"We are developing SB-509 as a first-in-class, disease modifying drug to protect and restore the function of damaged nerves," stated Edward Lanphier, Sangamo's president and chief executive officer. "To this end we have assessed the effect of SB-509 in a range of degenerative neurologic conditions and obtained positive clinical data in diabetic neuropathy and ALS, as well as preclinical data in models of spinal cord injury, stroke and traumatic brain injury. In addition to demonstrating safety, this clinical trial of SB-509 in ALS has provided us with valuable information for the design of future studies in this population in terms of potential end-points and dosing distribution and frequency. As we have previously reported, our clinical development resources are currently focused on our most advanced clinical program in this area, a Phase 2b study (SB-509-901) in subjects with moderate severity diabetic neuropathy from which we expect to have data in the fourth quarter of 2011.

WKN A0ESX5 für Minco Silver Corp, dabei Trendkanal beobachten. Es ist durchaus möglich das die Aktie zwischen 3,50 und 4,00 Euro nochmals zurückkommt. Wurde bei 3,70 empfohlen. Wenn der Trendkanal anhält ,dann sieht es für 2011 recht gut aus.

Sangamo`s nächste Hürde liegt bei 4,80 die nachhaltig durchbrochen werden muss.
Dann kann es schnell Richtung 7,00 laufen, wo meines Erachtens die nächste Wiederstandslinie liegt. Andersrum kann es auch Richtung 3,20 gehen. Es liegt daran welche Nachrichten demnächst Sangamo liefern wird.

man liest generel wenig von der firma aber steht ja auch im börsenbrief .
auf jedenfall sollte es baldigst anziehn werd ich noch mal nachlegen .
und wenn ich euch noch was sagen darf schaut mal bei wch888 ist wacker chemie kenn mich dort aus weill ich dort beschäftigt bin .

ich denke wir sehen hier noch viel viel höhrer kurze ihr könnt alle aussteigen bin erst 5,45 cent, eingestiegen, ich bleibe minimum 4 jahre bei dem wert... ich möchte endlich das grosse loss gesogen haben. warom bleibe ich solange bei diesem wert weil bei NOVO-NORDISK  vor 3 jahren von 10 euro bis jetz fast 90 Euro gestigen ist, bei solche werten  muss mann eben geduld haben...

Obwohl seit rund 15 Jahren Tausende so genannter Gentherapien in klinischen Versuchen getestet wurden, ist der Erfolg bislang ausgeblieben. Das könnte sich nun ändern: Das Startup-Unternehmen Sangamo hat eine Technik entwickelt, mit der defekte menschliche Gene gezielt repariert werden können.

Die meisten der Forscher, die sich Gentherapeuten nennen, haben sich in den vergangenen Jahren in Wirklichkeit eher als Genschmuggler betätigt: Sie versuchen, zusätzlich zum kaputten Gen eine intakte Version in die Zelle einzuschleusen – in der Hoffnung, dass diese sich durchsetzt. Doch häufig geht das kostbare Schmuggelgut auf dem Weg in die Zelle einfach verloren. In mindestens zwei Fällen endete der Austauschversuch sogar tödlich: So starb 1999 ein 18-Jähriger an einer Überdosis von Viren, mit denen ein Gen in seine Zellen eingeschleust werden sollte. Und bei dem zunächst als Erfolg gefeierten Versuch, eine Gruppe von Kindern mit einer erblich bedingten Immunschwäche mittels Gentherapie zu heilen, erkrankten drei der Kinder an Blutkrebs – ausgelöst durch das heilversprechende Ersatzgen, das im Erbgut der Patienten zufällig ein Krebsgen eingeschaltet hatte. Zwei der Kinder konnten geheilt werden, eines starb an diesen Nebenwirkungen.

Seither war die Gentherapie-Szene im Schock erstarrt – bis die Sangamo-Forscher im April 2005 an derselben Krankheit, der SCID (Severe Combined Immunodeficiency), zeigen konnten, dass sich mit ihrer Technik solche Risiken vermeiden lassen. Dabei kommen Proteine zum Einsatz, die selbsttätig die fehlerhafte DNA-Stelle ansteuern und reparieren.

Entdeckt hat diese Proteine der britische Forscher Aaron Klug – "Zinkfinger" nannte der Wissenschaftler seine Entdeckung, denn die kleinen Eiweiße hatten eigentümliche, fingerähnliche Ausstülpungen, die an der Basis durch ein Zink-Atom in Form gehalten wurden. Damit können sie buchstäblich nach DNA greifen und sie abtasten, um zu überprüfen, aus welchen Bausteinen sie besteht.

Ob die neue Technik, bei der keine Gene mehr unkontrolliert ins Erbgut eingebaut werden, wirklich sicherer ist als konventionelle Gentherapien, muss sich allerdings erst noch zeigen. Doch der Erfolg im Reagenzglas spornt die Sangamo-Forscher an, weiterzumachen. Neben Erbkrankheiten wie Sichelzellanämie und ß-Thalassämie, die Blutzellen betreffen, wollen sie auch Aids bekämpfen. Dabei sollen Zinkfinger ein Gen in den Blutzellen zerstören, das die HI-Viren für die Infektion der Abwehrzellen benötigen. Ebenfalls noch 2006 will Sangamo mit den Herzkreislaufspezialisten der US-Pharmafirma Edwards LifeSciences die Ergebnisse der ersten klinischen Studien präsentieren, in denen die Zinkfinger-Technologie zur Behandlung von Gefäßerkrankungen eingesetzt wird.

Zinc finger nucleases (ZFNs) are an extremely powerful tool for performing targeted genomic manipulation in a variety of cell types from Drosophila to humans. ZFNs consist of an engineered DNA-binding zinc finger domain linked to a non-specific endonuclease domain and can introduce double-stranded breaks (DSBs) that stimulate both homologous and non-homologous recombination, processes that can be harnessed to perform genomic manipulation. The capability to alter any genomic locus of interest will have tremendous potential in both research and gene therapy applications.

Targeted analysis of proteins on a
broad scale with mass spectrometry is
becoming a reality.
Based on rapid methodological developments
in 2009, last year we selected targeted
proteomics analysis by mass spectrometry
as a Method to Watch. Progress in 2010
continued at a quickening pace.
The targeted proteomics approach differs
fundamentally from the more familiar ‘shotgun’
approach in which the spectra generated
from all detectable proteins in a sample
are interpreted by database searching. In a
targeted analysis, the mass spectrometer
is programmed to analyze a preselected
group of proteins. This can be achieved
using a technology called selected reaction
monitoring (SRM; also referred to as multiple
reaction monitoring, MRM), whereby
assays are developed on a triple quadrupole
instrument to detect fragment-ion signals
arising from unique diagnostic peptides
representing each of the targeted proteins.
The SRM approach has proved to be
highly sensitive, quantitatively accurate and
highly reproducible. Compared to shotgun
proteomics (still largely the domain
of experts), protein detection is relatively
rapid and straightforward with SRM.
Though not yet quite as sensitive as immunological
assays, SRM has the clear edge for
multiplexed detection.
The main bottleneck to applying this
technology on a broad scale has been the
difficulty of generating high-quality SRM
assays. Over the last couple of years, methods
for selecting appropriate diagnostic peptides
and for generating SRM assays in high
throughput using crude peptides have been
developed. These developments have allowed
Ruedi Aebersold’s group to greatly expand
their SRM Atlas database (Nat. Methods 5,
913–914; 2008); as a result of an intensified
joint effort with Rob Moritz, this resource
will soon feature SRM assays for about 95%
of proteins from humans and yeast, and
about 55% of mouse proteins.
As the availability of SRM assays grows, so
will the biological applications of the technology.
Many researchers see great potential
in applying SRM for biomarker validation
and systems biology studies. Recently, SRM
was used to validate computationally predicted
microRNA targets in worms (Nat.
Methods 7, 837–842; 2010). This work likely
represents just a taste of interesting applications

improved design and efficiency will
become widely used.
The ability to make controlled changes in the
genome is a very powerful tool. Several technologies
exist for this purpose, but most have
drawbacks. For instance, transgenes introduced
into mammalian cells with viruses
are subject to position effects. Homologous
recombination does not work well in some
systems, such as human cells.
Zinc-finger nucleases (ZFNs) can be used
to create targeted double-stranded breaks
in the genome and have thus generated
much excitement. These enzymes consist
of a DNA-binding portion, which can in
principle be designed to target a particular
location in the genome, and a Fok1 nuclease
domain, which introduces the break. A
donor DNA can be exogenously supplied
to effect repair of the break by a homologydriven
process, resulting in gene replacement
with a desired sequence. Alternatively, mutations
are introduced
during an errorprone
repair
process.

systems: in mouse
and human cells, in
zebrafish, in plants,
in fruit flies and in the rat. ZFNs can be used
to create deletions of up to 15 megabase pairs
in human cells and have been used recently

stem cells, a system of tremendous interest
for genome engineering. No doubt the
application of these tools will continue.
One of the ongoing difficulties is that it is
not trivial to design ZFNs. Current methods
are either relatively inefficient at generating
enzymes that function well, or they are
laborious and require dedicated screening
systems. Many users depend on commercial
services, at relatively high cost. Methods
developers, however, continue to both
improve approaches to design ZFNs (for
instance, Nat. Methods 8, 67–69; 2011) and
to engineer enzymes with better properties
(for instance, Nat. Methods 8, 74–79; 2011).
An efficient and user-friendly method to
generate ZFNs that perform well is likely to
spread rapidly among researchers in many

ich lese hier ab und an mal mit. Noch wichtiger als einen steigenden Aktienkurs fände ich wenn Sangamo zum Wohle der Menschheit Erfolg hätte. Der Anfang Dezember angesprochene Börsenbrief... naja, mir ist der Verlag mit seinen "Chefredakteuren" etwas suspekt.

Einer davon behauptete doch tatsächlich dass auf der Autoshow LA zwei serienreife deutsche Limousinen mit E-Antrieb vorgestellt würden. Pusher, würde ich sagen.......
