Geron Reports on Successful Engraftment of Cardiomyocytes Derived from Human Embryonic Stem Cells

MENLO PARK, Calif.--(BUSINESS WIRE)--Aug. 20, 2003--Geron Corporation (Nasdaq:GERN - News) today reported the successful transplant of human cardiomyocytes (heart muscle cells) derived from human embryonic stem cells (hESCs) into animals. The study, conducted by Geron and Drs. Charles Murry and Michael Laflamme of the University of Washington School of Medicine, was presented by Dr. Murry at the American Heart Association meeting on Growth, Death and Regeneration in Myocardium.
In the study, hESC-derived cardiomyocytes were transplanted by direct injection into the myocardium (the muscular wall of the heart) of healthy rats. Histological examination of the rats at 1, 7, 14 and 28 days after transplant showed that the cardiomyocytes had successfully engrafted in each of the animals. Chromosomal analysis at each time point verified that the engrafted cells were human cells. Moreover, the grafts progressively matured in situ over the 28-day observation period, forming homogeneous tissue containing human cells that strongly expressed molecular markers of cardiomyocytes, including multiple cardiac-specific proteins. The mature grafts did not express markers specific to non-cardiac cells. Importantly, and in contrast to prior studies using mouse or rat cells, the cardiomyocyte grafts were highly proliferative in situ, with up to 15% of the grafted cells exhibiting markers of cell division at 28 days.

"These results are very exciting," commented Thomas B. Okarma, Ph.D., M.D., Geron's president and chief executive officer. "Not only are the cardiomyocytes engrafting efficiently, but they are maturing and expanding in vivo at the site of the graft. These results exemplify one of the basic advantages of using hESC-derived cells for tissue engineering -- namely their ability to re-create the natural developmental biology of new tissue formation."

The animals in this study had healthy hearts, so there was no opportunity to evaluate the effect of the transplant on heart function (e.g., the amount of blood that the heart can pump). Geron and collaborators are proceeding with studies involving animals with induced myocardial infarctions (death of heart tissue due to lack of blood supply), as a model of human heart disease.

Geron`s Duke cancer vaccine clinical Phase I/II trials (Stimulates the immune system to kill tumor/cancer cells:
1. Phase I/II, not just Phase I

Geron (WKN: 902213, US: GERN) steigen vorbörslich um 12.16% auf $8.30, nachdem das Unternehmen ein Patent für ein Telomerase-Gen erhielt. Dies löste eine Meilensteinzahlung von Novartis aus, hieß es. Geron hatte das Gen an Novartis lizenziert.

Geron Receives U.S. Patent for Human Telomerase Promoter Triggering Milestone Payment

MENLO PARK, Calif.--(BUSINESS WIRE)--Aug. 26, 2003--Geron Corporation (Nasdaq:GERN - News) announced today that it has been granted U.S. Patent No. 6,610,839, by the U.S. Patent and Trademark Office, with claims covering the promoter that regulates expression of the human telomerase reverse transcriptase gene (the "hTERT promoter"). Geron had non-exclusively licensed the hTERT promoter to Genetic Therapy, Inc. ("GTI"), a subsidiary of Novartis AG, for use in the development of viruses that are designed to target cancer cells. Cell Genesys, Inc. (Nasdaq:CEGE - News) recently acquired rights to the GTI product portfolio and intellectual property in the field of oncolytic viruses, including the hTERT promoter licensed non-exclusively from Geron. This granted patent triggers a milestone payment.

"This U.S. patent for the hTERT promoter is significant because pre-clinical animal data reported by us, GTI and others, are confirming that the promoter can confer both selective expression of effector genes and selective viral replication in cancer cells across a wide range of tumor types," noted David J. Earp, J.D., Ph.D., Geron's vice president of Intellectual Property. "These data suggest that the promoter can be used therapeutically to drive anti-cancer genes in cancer-killing viruses."

In June, 2003, GTI presented data from its work in oncolytic viruses at the annual American Society for Gene Therapy (ASGT) conference, held in Washington D.C. The studies used an oncolytic virus in which two genes essential for replication of the virus were controlled by separate tumor specific promoters -- the first by the E2F-1 promoter (primarily active in Rb-pathway defective cancer cells) and the second by the hTERT promoter. Based on studies with mice carrying human liver and prostate cancers, GTI reported that a single intravenous injection of the virus led to significant anti-tumor efficacy and improved survival, without significant toxicity. Moreover, the combination of the oncolytic virus with widely used chemotherapy drugs was synergistic in human tumor cell cultures of hepatocellular, prostate, small cell lung and colon carcinomas. Continuing the synergy studies in an in vivo liver cancer model, GTI reported that significantly improved anti-tumor efficacy was observed when the oncolytic virus was combined with doxorubicin, compared to either treatment alone.

We look forward now to Cell Genesys' efforts to continue the development of this product."

The telomerase enzyme must be turned on for cancer cells to replicate indefinitely and enable tumor growth and metastasis. Geron's proprietary telomerase technology is now the foundation for the development of three distinct approaches to treating cancer. Geron's telomerase inhibitors, GRN163 and GRN719, drugs that block the telomerase enzyme and send cancer cells into crisis, are in pre-clinical testing. Geron's cancer vaccine, which induces an immune response specific to telomerase-bearing cancer cells, is in a phase I/II clinical trial for hormone-refractory metastatic prostate cancer at Duke University. Finally, Cell Genesys will continue the development of cancer-killing oncolytic viruses that employ the hTERT promoter to drive selective expression of a gene required for viral replication in cancer cells.

Leider habe ich mich zum Zocken verleiten lassen und bei den Chinesen (IsteelAsia)investiert. Sollte ein schneller Zock werden und wollte dann umzwitschen. Leider schiefgegengen. Vielleicht trenne ich mich aber von QSC und kauf mir noch welche
Ich glaube nicht das Geron mir wegläuft da dass IND für GNR 163 erstmal auf mitte-ende nächsten Jahres vertagt wurde

Mein Kursziel auf zehn bis 15 Jahre beträgt dausend (Gründe gibt es genug wenn enttäuschungen (Klinische Zulassungskriterien, mögliche Konkurenzprodukte,etc.) ausbleiben sollten)

WSR: Good Day from Wall Street. This is LaVon ..., the senior analyst with Wall Street Reporter. Today our guest is Dr. Thomas Okarma, President and Chief Executive Officer of Geron Corporation, trading on Nasdaq, ticker symbol GERN and headquarters in Menlo Park, California. Good afternoon Dr. Okarma and welcome to Wall Street Reporter.

DR. O: Good afternoon LaVon. Thank you.

WSR: Thank you for joining us today. For our audiences who might not be familiar with the company, why don`t we start with a brief history and overview of Geron Corporation.

DR. O: Sure. The company was founded about 10 years ago and the founding notion was to try to understand the basic biology of diseases associated with aging and how that the actual process of aging itself contributed to those diseases. That notion quickly became formatted into what became the telomerase platform. Telomerase is an enzyme that adds DNA to the ends of chromosomes - that are called telomeres - and by doing so enables a cell to live forever. Telomerase has many roles in aging as well because the absence of the enzyme telomerase in our normal cells enables cells to become older as their telomeres shorten without the enzyme to add DNA back. On the cancer side of disease, telomerase is abnormally upregulated which is why cancer cells are immortal. The other point is that there are a small number of normal cells in our bodies called stem cells, like cells in bone marrow that make blood, which can express telomerase which is why they live for a relatively long time and are able to continue to repopulate, tissues that turn over, like blood or skin or the gut. So, the telomerase platform is a very important fundamental biological event that impacts cancer and aging.

What we`ve done with the platform is extract elements of it and format them into testable products, some of which are now in the clinic in cancer, others of which are a year or two away from testing in humans based on human embryonic stem cells. So let me perhaps give an overview of those programs.

The most advanced program on the cancer side of our company that is in Phase I/II trials at Duke University is a cancer therapeutic vaccine. We have learned how to turn the immune response on against telomerase which it turns out is expressed on the surface of cancer cells – all cancer cells. And we`ve developed with another company called Merix Biosciences a way to stimulate anyone`s immune system regardless of their tissue type against their own cancer using telomerase as an antigen because telomerase is expressed in everyone`s cancer type. So what we are experimenting on now are patients with prostate cancer and in April we announced the preliminary results of the first half of the trial which demonstrated that all the patients given the vaccine responded to it rather aggressively, meaning we were able to turn on their immune systems against telomerase and we demonstrated in all the patients who had elevated circulating levels of prostate cancer cells in their blood that the vaccine lowered those levels from six fold to a thousand fold, a very dramatic result, and that result, the elimination of metastatasizing cells, is the objective of the trial. Now, these data were reported -- there were absolutely no side effects – we were very impressed and pleased by these preliminary results. We have now gone on to enroll 12 more patients in the so-called high-dose group and we haven`t yet announced any of the results there, they`re still pending, but I can tell you that the high dose group is doing even better than the low-dose group that we reported on in April. So we will probably have some news to tell about that completed trial toward the end of the year. So the importance of this is that for us it validates the telomerase platform in cancer. If you target telomerase, you get no side effects and you get very specific activity against the tumor.

The next program in cancer, also based on telomerase, is an oncolytic virus. This is a virus that we have genetically engineered to make it dependent upon the enzyme telomerase in order for it to replicate and kill the cell in which it`s replicating. In other words, we`ve made a guided missile, a virus, that only replicates and kills telomerase positive tumor cells. We`ve licensed that to GTI/Novartis who has done a terrific job in moving that technology forward, and they have just, as you may recall, licensed that to Cell Genesys who had also been looking at our telomerase promoter for their oncolytic virus program. So Cell Genesys will now take over the development of that product from GTI/Novartis and we`re very pleased actually with how that`s turning out. So that program – I can`t tell you when it will enter the clinic, obviously Cell Genesys has to kind of catch up with what GTI/Novartis was doing, but we expect to be able to announce something about that fairly soon

The third and probably most important element of the oncology program is our telomerase inhibitor drug, and this is, I believe, will be Geron`s home run in cancer. We have for l0 years been working on a drug to shut down the enzyme telomerase and to shut it down in a very elegant, specific and powerful way. And we have now discovered two compounds called GRN 163 and GRN 719 which are absolutely specific and potent inhibitors of the enzyme telomerase. These compounds have demonstrated tumor killing in 7 or 8 different in vivo models and in virtually all of the major human cancers in vitro. And they do this without any toxicity at the therapeutic dose to normal cells, which is a new paradigm in cancer treatment. Most cancer drugs are nonspecific – they attack pathways that are in normal cells as well as in cancer cells. This drug only attacks the telomerase in a tumor cell, so it`s elegantly specific and powerful and we and others have demonstrated not only potent activity in animals of this drug but a very impressive synergy between our drug and other chemotherapy drugs. And the importance there of course is that our drug does not add to the toxicity of the other drug used in combination. So we are in advanced preclinical studies with these two compounds. They are essentially the same sequence. In terms of the composition of the drug, the 719 molecule is lipidated, which means it gets into cells better and is actually more potent. So we`re now doing head-to-head comparisons of the two compounds to pick one to move into the clinic.

So that`s the cancer side of Geron, different ways to use the enzyme telomerase which is still today the world`s only validated universal cancer target to fight tumors in man.

Now the other side of Geron is also based on telomerase, but it`s based on the other element of telomerase, which confers genetic stability onto long lived cells. This is the stem cell story. The reason why our skin and our gut and our blood cells are constantly turned over and maintained during our lifespan is that the cells that make skin and gut and blood can upregulate telomerase. Telomerase does not cause cancer; telomerase is simply an enzyme that enables a cell which makes it to live forever. And that`s how stem cells use it - to enable them to live forever. We`ve discovered that the embryonic stem cell, which we derived in 1998, also expresses telomerase. It turns out that in normal human development all cells in the developing fetus in the first trimester are telomerase positive. It`s not til later in in utero development that the telomerase is turned off, to reside only in those adult stem cells we talked about. So it turns out that the embryonic stem cell, because of its telomerase expression, is immortal. We have had these cells growing continuously in culture now for years. Hundreds of population doublings of these cells and they remain absolutely constant in their properties. There is no other cell in the universe that has these characteristics. The importance of that is it enables us to scale up the therapeutic cells that we make from embryonic stem cells in the same way that companies make protein drugs or monoclonal antibodies and that has never been possible before in cell therapy.


Now the other feature that makes embryonic stem cells important is that they can be turned into all of the differentiated cells in our bodies. That`s why these cells are so powerful. So we`ve now learned how to make many differentiated therapeutic cells and are in animal studies now with four of them. So we can make, for example, oligodendrocytes which are neural cells that line and wrap around or insulate spinal nerves and we`ve demonstrated with collaborators at UC Irvine that when we inject these human glial cells into the spinal cord of injured animals they show dramatic functional recovery to their injury and when we look at the site of the injury under the microscope we find that the human cells have actually repaired and re-engineered what otherwise would have been permanent damage to the spinal cord. This is very exciting news. And we expect the oligodendrocyte cell type to be the first cell type we put into the clinic. If all goes well and perfectly, this could be the end of next year or the beginning of the year after, and that would establish the proof of concept in man of embryonic stem cells first and foremost. Now, behind the oligodendrocytes are other cell types equally exciting.

We`ve derived dopaminergic neurons to treat Parkinson`s Disease and we are working on implanting them in animal models of Parkinson`s and we`ve demonstrated robust engraftment. We are still optimizing the process to try to demonstrate that we can get recovery of the Parkinsonian-like symptoms in these animal models. We`ve not yet demonstrated that, but we`re getting very robust engraftment of the human dopaminergic neurons in the animal models. We have also derived hematopoietic stem cells, the cells that make blood and these are in animal studies and are engrafting as well. The purpose of the hematopoietic cell is not so much for bone marrow transplant, but it`s a way for us to prevent immune rejection of any of these transplanted cells. That goes back to the bone marrow transplant story, and the, the details are very simple. If I were to give you a low dose of stem cells that form blood made from let`s say embryonic Stem Cell Line A, that will tolerize your immune system to any other cell type that we make from Stem Cell Line A. So it`s a way for us to give you the therapeutic cell type, the neurons, the oligodendrocytes, whatever, without your needing immunosuppresive drugs and that`s important because some of the applications, like diabetes and heart failure that we`ll talk about in a moment would otherwise require the patient to take immune suppressive drugs for life. They`re expensive and they`re toxic. So it`s important that we have a way to circumvent that requirement and we`ve now accomplished that in animals.

Another cell type that is in animal studies, again very exciting, are cardiomyocytes, heart muscle cells. We just reported a few days ago on a collaboration where we`ve demonstrated that our human heart muscle cells engraft in the animal`s heart and they mature in situ; in other words, over the three weeks, four weeks, that we followed the animals, you actually see the graft maturing and forming normal junctions between the cardiomyocytes that will insure normal cardiac electrical and force transmission. We`ve already shown that these human cardiomyocytes respond to cardioactive drugs normally. So when we move into human clinical trials and put these cells into the hearts of patients with heart failure, we know that the drugs they`re also on will be affecting their own heart cells the same way they`ll be affecting the new ones we put in.

We`ve now derived the islet cell, the cell that makes insulin, for diabetes. We have not got it scaled yet, it`s not yet in animal studies, but it is a glucose responsive cell that appears to make insulin in response to glucose. This example in diabetes is probably the most exciting because the proof of concept of cell therapy in diabetes has already been demonstrated by the Edmonton Protocol in Canada where cataveric islets are placed into the livers of patients with diabetes where they set up sort of a pancreatic like islet and can respond to glucose and make insulin. Of course there aren`t enough cadavers to service the population, so, again, the scalability of embryonic stem cells is a step toward solving that issue. So we`re very excited by that recent development.

We`ve also made bone forming cells for osteoporosis and nonunion fractures and we`re working hard on making cartilage which will be of course useful for patients with various forms of arthritis.

So in general the platform of embryonic stem cells is a major step forward to actually achieve the objectives of what`s called regenerative medicine: learning how to do what drugs cannot do, which is to actually repair and re-engineer the tissue in various organs: brain, spinal cord, pancreas, heart, and so on, that are damaged by a chronic disease and which no drug can restore the function of. So for the first time we see ourselves entering the space of being able to efficiently in terms of low cost of goods and scalability repair tissues by means of cell therapy.

We have lines that are now qualified for human use. They have passed all of the FDA`s required tests to demonstrate their safety in man and we`re now working very hard on scaling up and validating these processes because the product we take to the FDA in these cases will be differentiated cells, scalably made from embryonic stem cells that will be stored frozen, ready for off the shelf use for patient trials. So that`s the spectrum of Geron`s activities.

Oncology programs that are in the clinic or near the clinic which are looking extremely exciting. A new paradigm for cancer treatment, very elegant and specific, based on telomerase expression in tumor cells. And behind that, a much broader and at least equally dramatic opportunity in regenerative medicine where each of the cell types we`ve been talking about addresses an enormous market opportunity. Enormous whether it`s looked at in terms of the incidence or prevalence of the disease or in terms of the actual economics involved given that these are probably one or two times at most therapies and will be highly priced. And the scalability of the manufacturing process enables us for the first time to make these products with the same economies of scale as monoclonal antibodies or as biological drugs.

So the future for us I think is extraordinarily bright. We have segued from a company that discovers and patents its technology platforms to a company that is now testing and developing and doing preclinical GLP work on products that we have developed from our platforms. And if you pay attention to our press releases, I think it documents the fact that we have moved from a discovery environment to a development environment, where the patents that are now issuing for us aren`t on the basic technology anymore, they`re actually specifically covering the cell types we make or the drugs that we`re moving into the clinic or the elements that we use in our vaccines or oncolytic virus products. So, we`ve turned the corner I think from discovery to development, and I think the take home message is to stay tuned for continued news as we move these programs into the clinic and beyond.

WSR: Dr. Okarma, as we look at your current cash positioning, do you have enough funds and resources available to support and move your programs?

DR O: Sure. I mean that`s a good question. As you know, we have done the hard things. We have done two reductions in force, one last year and one at the beginning of this year. Those are painful things that you don`t want to do unless you have to. We did it to conserve our burn rate and we were able to do that because we were able to retain the employees who were on the development side and unfortunately let go of the employees who were on the discovery side. So we have not lost our momentum and have certainly conserved, reduced our burn rate. We raised some money in April. We have funds at the current burn rate through ‘05. We will of course need to raise more money at an opportunistic time and we are always ready to do that, so we do not have the funds today to take all of these programs through IND filing. Of course. But what we usually do is keep two to three years running room in the bank which we have today and we will continue to do that strategy.

WSR: As we look at your stock performance over the last six months, do you feel the investment community is starting to recognize the potential?

DR. O: Well I think, yes is the short answer. I think there was a lot of concern with the second reduction in force as to whether Geron was in, you know, quote "trouble." I think the news flow since that time has reassured people that we did the hard thing in terms of internal management, but did not do anything to alter continued productivity and progress toward the clinic. I think as the portfolio matures and particularly as more data becomes publicly available for the vaccine, people will begin, investors will begin to recognize that this is now a product company and that the promise of telomerase on both cancer and the regenerative medicine side of our company is going to be realized and realized in a very dramatic way, dramatic both for patients with these diseases and for investors who have positioned themselves in the company at a time when the stock price is where it is now.

WSR: Dr. Okarma, lastly, what are some of the key milestones or benchmarks that our investors should keep an eye on over the next 6 to 12 months?

DR. O: Sure. I think first will be the announcement of the completion of the Phase I/II trial at Duke. We hope that that will be available by the end of the year. Nearly all the patients are enrolled , but these are patients who must be followed for months after the last vaccine, so it`s simply a matter of data accrual and when all of the important variables are intact so that we can give a complete story - we hope that will be within the next 4 to 6 months. I think a little more distantly, hopefully within the next 12 months will be an IND filing for the telomerase inhibitor - that will be a major watershed event given the upside for that cancer. We would hope to be able to talk also in that time frame more specifically about the plan for the oncolytic virus now in the hands of Cell Genesys. I don`t think it will be likely that we will file an IND for spinal cord within 12 months, but it may well be within 18. There`s some scale up issues to do and some toxicity that we have to do in other animal species. We are optimistic though that in the 12 to 18 month time frame the oligodendrocyte spinal cord injury trial will begin. So those are the three really big ones. We will continue to advance our patent portfolio. We have an impenetrable estate on the telomerase side and are beginning to accumulate that patent estate on the stem cell side. And I should mention that the IP protection for the embryonic stem cell portfolio includes composition of matter claims for the cell types, not just methods of how to make them. That puts us in a very dominant position worldwide against competitors.

steiler Anstieg auf knapp 20 Dollar. Das wäre für mich der Zeitpunkt zum vorübergehenden Ausstieg. Ansonsten befürchte ich, dass die Sache so vor sich hindümpelt und man immer wieder mal wegen einer vielleicht nur vermeintlich schlechten Meldung (als Beispiel siehe Protein Design Labs) um 25 Prozent runtergeprügelt wird. Die werden dann zwar wieder abgebaut, aber es nervt. Also, sorgt für einen rasanten Anstieg und die Rettung meines Weihnachtsfestes. Ich will dieses Jahr nämlich eine goldene Gans.

Gerons Patenterfolge nehmen kein Ende

Die Aktie der Geron Corporation (Nasdaq: GERN) bereitete den Investoren gestern wieder große Freude, leider konnten von der guten Nachricht vom Erhalt eines neuen US-Patents nur die bereits engagierten Geron Anleger profitieren, denn Geron legte bereits im vorbörslichen Handel deutlich zu. Das an Geron erteilte US-Patent schützt Ansprüche am Promotor des Gens für das Enzym Telomerase Reverse Transkriptase, den Geron an Genetic Therapy, eine Division von Novartis (Nyse: NVS), lizenziert hat. Der Promotor ist ein Genabschnitt, der sich am Anfang des Gens befindet und für die Ausprägung des Gens und damit für die Menge des Enzyms Telomerase Reverse Transkriptase verantwortlich ist. Novartis will mit dem Promotor Viren entwickeln, die sich zum Kampf gegen Krebszellen eignen.

Die Ausgabe des Patents an Geron löst eine Meilensteinzahlung durch den Lizenznehmer an Geron aus. Über die Höhe der Meilensteinzahlung wollte Geron keine Auskunft geben. Erst kürzlich hat Cell Genesys das Produktportfolio sowie das gesamte intellektuelle Eigentum im Hinblick auf die von Genetic Therapy entwickelten onkolytischen Viren erworben. Dies schließt auch den von Geron lizenzierten Promotor für das genannte Gen mit ein. Den in der Vorbörse errungenen Kursgewinn von 12 Prozent konnte Geron leider nicht halten, allerdings konnten sich die Aktionäre immer noch über einen Aufschlag von 7,3 Prozent freuen.

Mit diesem erneuten Erfolg setzt Geron seine bereits vor mehreren Wochen begonnene Glückssträhne erfolgreich fort. Die Zukunft von Geron sieht gegenwärtig recht rosig aus, wer zu den glücklichen Besitzern dieses Wertes gehört sollte ihn noch eine zeitlang hüten, denn Geron dürfte noch einiges mehr zu bieten haben.

War mal eine Zeit lang in Trinity Biotech investiert. Die haben auch so einen Schnelltest auf Aids.

Calypte hat natürlich nun einen Vorteil aber ob das alles so wird wie es aussieht!
Ich glaube nicht, auch wenn das AIDS- Problem ein Problem der armen Länder ist so zählt ein Menschenleben auch gerade deshalb dort nichts. Leider ist es so! Weniger Menschen weniger Kosten!! Man hat einfach nicht das Geld um eine menschengerechte Gesundheitspolitik zu gestalten. Kann sein das ich hier verkehrt liege und mir es leid tut nicht in Calypte investiert zu haben aber ich bleibe dabei, nicht in diesen Wert investieren!

Calypte kratzt doch noch die Kurve !

Vor geraumer Zeit hatten wir vom Biotech-Experten (www.Biotech-Experte.de) ein Unternehmen aufgestöbert, dass uns aufgrund seiner Produkte in Kombination mit seiner Bewertung als interessant erschien. Trotz der Tatsache, dass Calypte Biomedical mit dem ersten durch die FDA zugelassenen HIV-Urintest ein hervorragendes Produkt besaß, war die Bewertung des Papiers gigantisch niedrig, was uns dazu veranlasste, dem Unternehmen einmal etwas genauer auf den Zahn zu fühlen. Nicht lange mussten wir suchen, da fanden wir auch schon den Grund für die unserer Meinung so niedrige Bewertung. Calypte war in Finanznöte geraten und stand kurz vor der Insolvenz. Doch wir hatten einen guten Riecher und empfahlen den Wert als spekulatives Investment und kurz darauf kam tatsächlich mit einem Privatinvestor der rettende Anker für Calypte. Der Kurs des Pennystocks zog damals deutlich an, doch im letzten Quartal hatte Calypte erneut mit Chapter 11 gedroht, was wiederholt zum Kurseinbruch der Aktie geführt hatte.

Ich muss gestehen, so richtig daran geglaubt, dass sich Calypte doch noch einmal aufrappeln könnte haben wir ja nicht, dennoch waren wir uns sicher, dass man ein effektives und sicheres Testverfahren für HIV, das zudem noch die FDA Zulassung besaß, nicht verkommen lassen würde. Finanzspritze oder Übernahme das war lediglich die Frage bei Calypte, die wir uns immer wieder stellten. Und wie sich in den vergangenen Tagen gezeigt hat, hatten wir gar nicht so unrecht mit unserer Einschätzung, denn Calypte wird nun wohl doch weiter bestehen bleiben. Fast beängstigend waren die "fast" grundlosen Kursgewinne der vergangenen Tage, am Donnerstag plus 69 Prozent, am Freitag plus 56 Prozent und das bei einem gigantischen Handelsvolumen von über 20 Millionen Aktien. Gewöhnlich werden von Calypte im Durchschnitt nicht mehr als 700.000 Aktien gehandelt. Doch da an der Börse nichts ohne Grund geschieht, waren wir vom Biotech-Experten schon nach dem ersten Anstieg skeptisch und haben den Braten bereits gerochen.

Und siehe da, der Dienstag brachte dann endlich des Rätsels Lösung, das wohl einigen Investoren schon seit einigen Tagen bekannt gewesen sein muss, wofür der rasante Anstieg des Kurses bei Calypte in den vergangenen Tagen spricht. In einer Pressemitteilung wurde gestern dann endlich allen Investoren die freudige Kunde von einem Investment in Höhe von 10 Millionen USD durch die Marr Group überbracht, worauf Calypte erneut um mehr als 60 Prozent gen Norden zog und damit wieder einmal der schimmerndste Stern im US-Biotechsektor war. Mit den bereits am 31. Juli erhaltenen 2,5 Millionen USD durch Marr Technologies Limited, einem Mitglied der englischen Marr Gruppe, besitzt Calypte nun 12,5 Millionen USD in bar. Genügend also, um seine künftigen Pläne, die weitere Verbreitung des HIV-Urintests, in die Realität umzusetzen.

Die Marr Gruppe verfügt über eine Vielzahl von verschiedenen Investments auf weltweiter Basis und ist vor allem auf dem chinesischen Markt aktiv. China, wo Calypte seine Tests schon an zwei Distributoren ausliefert, steht gerade am Anfang einer AIDS Epidemie. Das Land hat bereits international um Hilfe gebeten, um eine Epidemie wie in Afrika möglicherweise noch abzuwenden. Eine landesweites Screening auf das HI-Virus wird sich also kaum umgehen lassen und dafür ist der Urintest von Calypte, der keine Ansteckungsgefahr in sich birgt und selbst von medizinischen Laien durchgeführt werden kann, geradezu prädestiniert.

Die Entscheidung von Marr für ein zusätzliches Investment in Calypte kam aber nicht von ungefähr, sondern folgte einem Entscheid der WHO auf die weitere Evaluierung der Calypte HIV-1 Urintests zu verzichten. Die WHO ist der Meinung, die bisher erzielten Daten einer Phase I Studie sowie die bei der Anwendung ermittelten Daten werden für eine Entscheidung im Hinblick auf die Empfehlung des Test genügen. Im Rahmen dieser Vereinbarung wird Calypte nun ein Dossier abfassen, dass sämtliche bisherigen Daten beinhaltet und dies der WHO für ihre entgültige Stellungnahme zur Verfügung stellen. Sobald ein wohlwollender Bericht der WHO vorliegt, wovon man ausgehen kann, dürfte die Nachfrage nach Calyptes HIV-Tests deutlich steigen. Grund für diese Annahme ist die Tatsache, das viele Länder sich in ihrer HIV Politik nach den Empfehlungen der WHO richten. Damit ist Calypte nun wohl wieder auf dem richtigen Weg zu den alten Höchstständen. Derzeit sind zumindest keine weiteren Hürden in Sicht.

Habe Morphosys verkauft und bin wieder in Greon rein. Unter Strich hat mir die ganze Aktion 150 Euro gebracht. Leider ist Geron zu schnell angezogen und der ganze Gewinn dadurch zusammen geschmolzen

Warum Geron heute leider so stark anzog und mich meine Position in Morphosys zum auflösen veranlasste, ist diese Meldung.

Dendreon Gets Fast-Track Status for Prostate-Cancer Drug

SEATTLE -- Dendreon Corp. (NasdaqNM:DNDN - News) said the Food and Drug Administration (News - Websites) has granted fast-track status to its investigational prostate-cancer vaccine, Provenge.
Under the fast-track designation, the biotechnology company said Thursday that it will be able to enact a rolling submission of a potential Biologics License Application for the cancer vaccine. The company had planned to complete a BLA by early 2005.

The news pushed shares of Dendreon sharply higher Thursday. Near 11:30 a.m. EDT, Dendreon's shares were up $1.53, or 25%, to $7.76 on the Nasdaq National Market.

The purpose of fast-track status is to accelerate the development process for drugs that treat a disease that was previously untreatable.

Provenge is designed to stimulate a man's immune system against prostate cancer, the leading cancer diagnosed among men in the U.S.

"The designation of Provenge as a Fast Track program by the FDA validates its promise to address a serious unmet medical need for the treatment of advanced, asymptomatic androgen independent prostate cancer," said Dr. Mitchell H. Gold, Dendreon's chief executive, in a prepared statement.

In June, the FDA agreed to let a second late-stage trial of Provenge serve as the basis for a Biologics License Application for the vaccine.

Dendreon said its fast-track application was based on positive results from a Phase 3 trial that indicated the drug delayed the progression of prostate cancer.

The company is currently conducting a separate late-stage trial of Provenge that seeks to confirm the results of previous trials.






Geron hat die rechte an Dendreon auslizensiert und verdient über Meilensteinzahlungen ebenfalls was aber interessanter an der Sache ist Geron dass Geron ebenfalls einen Impfstoff in den klinischen Phasen hat der ähnlich wirkt. D.h. die FDA verteilt Fast trach auch für Teöomerase-Impfstoffe und Geron ist auf diesem Gebiet der Telomerase-impfstoffen und Hemmer führen.

Dendreon weist derzeit ein Kursplus von 40 % auf

DALLAS - Sept. 15, 2003 - Certain enzyme inhibitors may slow tumor
formation within weeks and could lead to treatments that retard or
prevent recurrences of cancers, researchers at UT Southwestern Medical
Center at Dallas have discovered.

Their findings appear in the current issue of the journal Cancer
Research.

The researchers sought to inhibit telomerase, an enzyme that maintains
telomeres - repeating sequences of DNA at the end of each chromosome
that are believed to function as a counting mechanism for cellular
aging. Telomerase prevents the shortening of the sequences of DNA that
occurs in normal cells as they age. The enzyme is found in most types of
tumor cells but not healthy cells, indicating telomerase inhibitors may
be a powerful new approach to chemotherapy.

Telomerase inhibition, however, has posed challenges for therapy. In
earlier studies, scientists have found that months of treatment with an
inhibitor are required before tumor growth could be expected to
significantly slow.

The UT Southwestern researchers treated cultured human tumor cells with
a unique compound that blocks telomerase activity, and the cell
proliferation slowed substantially after just a few weeks.

Further, prostate cancer cells treated with the inhibitor barely formed
tumors in mice and yielded very low levels of prostate specific antigen
(PSA), a marker associated with malignancy. Cells treated with a similar
compound that was not a telomerase inhibitor formed large tumors with
high PSA levels.

" Telomerase is widely appreciated as a promising target for therapy,"
said Dr. David Corey, professor of pharmacology and biochemistry and the
study`s senior author. " Our results suggest that if you can inhibit
telomerase in tumor cells and shorten telomeres, you will slow the
growth of tumors."

The researchers also discovered that when the telomerase inhibitor is
combined with standard cancer therapeutic agents carboplatin and
cisplatin, there are additional antiproliferative effects. Dr. Corey
said these results suggest a relatively small amount of telomere
shortening is sufficient to slow tumor growth, and telomerase inhibitors
are a useful therapeutic option, especially in combination with agents
already being used to treat patients.

" No one is suggesting telomerase inhibitors alone would cure cancer,
but in conjunction with standard therapy, they might help to slow or
prevent the recurrence of tumors after the initial cancer has been
removed through surgery, radiation or chemotherapy," Dr. Corey said.
" Since most patients die from the recurrence of cancer, effective
telomerase inhibitors could have a large impact on the treatment of many
different types of cancer."

A similar telomerase inhibitor currently is in advanced preclinical
trials with the Geron Corp. These new findings are likely to influence
how clinical trials are designed and interpreted and to provide more
support for pushing them forward, Dr. Corey said.

Dr. Zhi Chen, pharmacology postdoctoral researcher, and Dr. Kenneth
Koeneman, assistant professor of urology, also contributed to the study.

Telomerase verhindert den Shortening der Reihenfolgen von DNA, die in den normalen Zellen auftritt, während sie altern. Das Enzym wird in den meisten Arten Tumorzellen gefunden, aber die nicht gesunden Zellen, telomerasehemmnisse anzeigend können eine leistungsfähige neue Annäherung zur Chemotherapie sein. Hemmung Telomerase jedoch hat Herausforderungen für Therapie aufgeworfen. In den früheren Studien haben Wissenschaftler gefunden, daß Monate der Behandlung mit einem Hemmnis angefordert werden, bevor Tumorwachstum erwartet werden könnte, um erheblich zu verlangsamen. Weiter behandelten Prostatakrebszellen mit den gebildeten Tumoren des Hemmnisses kaum in den Mäusen und erbrachten sehr niedrige Niveaus des Prostataspezifischen Antigens (PSA), eine Markierung, die mit malignancy dazugehörig ist. Zellen behandelten mit einem ähnlichen Mittel, das nicht ein gebildete große Tumoren des telomerase Hemmnis mit hohen PSA-Niveaus war. " Telomerase wird weit als vielversprechendes Ziel für Therapie," geschätzt, sagte Dr. David Corey, Professor von Pharmakologie und von Biochemie und von älteren Autor der Studie. We wrote: "Geron, a former pick of ours back in May and then again in July, has been consolidating for the last 90 days. Underlying technicals indicate strong accumulation going on, as they are now at the highest levels in the last couple years. Overhead resistance at 9 1/2/-3/4 needs to be overcome before Geron can really move, but should that occur we expect a move to test the top of the trend channel as well as overhead price resistance in the 13-14 zone, my initial target. Secondary target is 18 ½-19, and longer-term target perhaps in the low-to-mid 20s. Underlying support is down around the 7 ½-8 zone, and beneath that around 6 ¾-7. But the overall pattern looks very strong."

Okay. Good afternoon. Like to introduce our next speaker, Dr. Thomas Okarma, President and CEO of Geron.

Dr. Okarma: Thank you, David, and thank you all for coming today. I know this won`t surprise you but I too will be making forward looking statements so we call your attention to the risk factors in our 10Q.

We have two stories to summarize for you today and I`m going to start with oncology, the cancer platform, which is really our near term value driver. And the take home message for you today in cancer is that we believe we`ve now clinically validated the telomerase target. You may know that telomerase remains today the only known universal cancer target, meaning, it`s expressed in all cancer cells and very rarely in normal cells. We have three therapeutic programs that address different elements of telomerase in cancer.

The most advanced is our Phase I/II trial in prostate cancer at Duke where we`re now showing and I`ll give you the data today, that telomerase vaccine, vaccination safely induces strong anti cancer immune responses in advanced cancer patients.

We have an oncolytic virus program as well, initially licensed to GTI/Novartis, now acquired by Cell Genesys where we have now shown in multiple animal studies dramatic efficacy and synergy with chemotherapy.

And lastly, our home run, the telomerase inhibitor drugs, 163 and the newer variant 163L. Literally, these compounds are effective in vitro against all the major human cancers and in animal models have been effective in all six models that we have thus far exposed these drugs to. So let`s go a little bit deeper into this, and of course recall that telomerase is the tap root of Geron and as such we have a controlling intellectual property estate on the platform and the products you`re about to hear.


The trial at Duke we`ve described in April. It is a two phase trial with a low dose group of 12 patients and a high dose group of 12. We reported some of the preliminary results back in April which I`ll now remind you of. First, all of the patients in the low dose group - all 12 of them - responded with vigorous immune responses specific to telomerase. All of these red bars are T cells that react specifically to telomerase in all of the patients, and this is significant because these are advanced cancer patients who are by definition immune suppressed. We also demonstrated in the first low dose group that all 7 of the patients who had elevated circulating levels of metastasizing prostate cancer cells had those levels reduced to zero concomitant with the vaccination. Some of these patients lost over a thousand fold of circulating prostate cancer tumor cells – no patient did not respond to this vaccine in that way.

The new data that I can share with you today is the beginning of the results from the second phase, the so-called high dose group who received six vaccines instead of three in the low dose group. And there are several points of importance to be made here. First the absolute level of T cells that we are generating in these subjects in the high dose group is between one and two percent of their total T cell pool; that`s never been achieved in cancer vaccination. That`s the level of T cell reactivity you see with measles or mumps vaccinations. Second, the duration of the immune response appears to be very long lived, well after the sixth vaccination and in fact the stabilizing level of T cell immunity in the high dose patients - at 13 weeks - is higher than the peak level we got in the low dose group where we demonstrated the efficacy with regard to circulating tumor cells. So we`re very excited that by simply adding three more injections of the same number of cells we`ve gone from a short term low but effective immune response to one that appears to be long lived. So stay tuned later in the year when we now, when we unveil the results of the rest of the high dose group and focus on clinical responses.

We have good surrogates now for immune response which is the skin rash these patients develop at the injection site. We have actually biopsied this delayed hypersensitivity reaction and shown it to contain anti telomerase T cells. So we now have a visual measure of the immune response.

And none of the patients in the low dose group or the high dose group have experienced a single adverse reaction.


Another piece of evidence on the vaccine side came to us indirectly. In another trial not sponsored by Geron at Duke in renal cell carcinoma when the patients were immunized with total tumor RNA we knew that they had responses, responses to laminen and specific antigens in renal cell carcinoma that had been known, what we didn`t know until we did the analysis though was that the dominant antigen in the total tumor RNA in these patients is in fact telomerase - and these patients are enjoying clinical responses.

So those two lines of evidence show that when you target telomerase you get effective anti tumor immunity and no side effects.

Let`s turn quickly to the oncolytic virus. This is a genetically engineered virus in which we control the replication of the virus with the telomerase promoter, the on/off switch. So the only way the virus can replicate and kill the cell it`s replicating in is if it`s in a telomerase positive tumor cell. And we and GTI/Novartis have published on the specificity of this virus before. So what I want to show you here is work also done at GTI that shows synergy of this virus with doxyrubicin. So here we`re measuring tumor volume, the spread or growth of the tumor, of a liver cancer in an animal. The virus alone is shown here in blue with a effect that is significant over control. Here is doxyrubicin by itself. At this dose slightly more potent than the virus by itself. But in these two lines we see the combination of that dose of virus and that dose of doxyrubicin, which is literally complete control of the tumor. Very dramatic synergy. And, again, because we control the replication of the virus elegantly to telomerase, there`s no adverse events in these animals. So we look forward to the continued development of this technology with Cell Genesys

Now the final entry in the oncology space we think are our home run compounds, GRN 163 and its lipidated cousin GRN 163L. Now it`s now agreed upon worldwide by basic scientists and oncologists itself, themselves, that, that cancer progression is inexorably linked to telomerase activity. It is still the only universally validated cancer target. We have worked for a long time to try to generate drugs that are as specific against telomerase as telomerase is specific to cancer to leverage that value of that target. And we have done that with these two compounds which are potent, specific, and absolutely nontoxic inhibitors of telomerase. We have literally demonstrated that 163 is active in vitro against all of the major human cancers in man shown here. We`ve begun now to do animal studies with some of these tumors, and these six cancers are effectively suppressed in vivo in animal models with GRN 163: brain cancer, prostate cancer, lymphoma, myeloma, liver cancer and cervical cancer, and I`ll show you a bit of that data today to give you a feel for how dramatic this compound may be clinically.

This is a picture of 163. A space occupying molecular model. It`s a 13 mer oligonucleotide, 13 building blocks. It is not an antisense molecule. It has no antisense activity whatsoever. It is a direct competitive inhibitor at the active site of telomerase and it is the only molecule in the human body to which this compound binds. It is effective both intratumorally, and I`ll show you that data in a moment, as well as parenterally. We are doing multiple GLP toxicology studies now, we have multiple manufacturing contracts in place and have IP protection not only for the compound but for the unique chemistry that makes it so potent and specific, as well as the target and issued IP for the clinical use of this compound to treat human patients with cancer. Now we`ve shown these data before, the intratumoral application of 163 in animals bearing a very malignant form of brain cancer and the results are quite striking. All the control animals are dead by day 43, 5 of 7 of the treated animals are alive and well at the time they are sacrificed by protocol at which time they are examined histologically for any evidence of residual tumor, and 5 of the 7 animals show zero tumor in their brain. Now that wouldn`t be so surprising if 163 were a toxic compound, but those same animals show absolutely no adverse reactions whatsoever. This is truly a new paradigm in chemotherapy for cancer: specificity, efficacy and the absence of toxicity.


Some new data that I`ll share with you today from Sloan Kettering here in New York in Malcolm Moore`s lab. This is disseminated multiple myeloma done in a way that the colors indicate the spread of the tumor in the animal - so blue color going to red is the presence of the multiple myeloma in the animal`s body. So here on the control animal, as you can see, dramatic spread of the cancer throughout the animal. Here is the maximally toxic--tolerated dose of doxyrubicin, the best compound to be used in this form of cancer and it`s actually somewhat worse than PBS control. In contrast, seven days of treatment with 163 systemically produces this dramatic result, colorometrically that you can see visually , the absence of blue which when quantified shows dramatic efficacy 30 days after the onset of, after the introduction of the cancer. So another example of the systemic potency of 163 as a stand-alone agent in a remarkably malignant tumor which outperforms doxyrubicin at its maximally tolerated dose, and, of course, we enjoy a substantial survival benefit which obviously is the exercise at the end of the game. Now 163L which we used to call 719 is the lipidated version of 163. It`s the same molecule, same chemistry, but we hang covalently a lipid molecule on one end of the drug. This has enormous impact and we are now doing head-to-head studies of 163 vs. 163L in tox, in efficacy, in synergy, in PK, so that we choose the best compound to move forward into the clinic. Clearly the drug has enhanced bioavailability, which I`ll show you, and potency. It also has manufacturing advantages. It`s less expensive to make and, of course, was designed for systemic administration. So here`s the kind of data we`re getting when we compare 163 head-to-head with 163L. Blue is PBS control, the pink is 163 and the green is 163L at a lower dose than 163 done in the same model. So what you see is a low dose of 163L is much more effective than a higher dose of 163 in inhibiting telomerase in the cells in the animal, in the, in the cancer. Also, the length of telomeres, telomere shortening, which is the mechanism of action, at the lower dose of 163 is more potent than the higher dose of – I`m sorry – the lower dose of 163L is more effective at shortening telomeres than the higher dose of 163. If we now look at the whole animal data, we see the same story. The lower dose of 163L is more potent in terms of tumor inhibition in the animal than the higher dose of 163. So those are the kind of data that we`re now getting that augur well for the clinical introduction of the lipidated form of this bio-molecule.


We are now in the toxicity phase of testing 163L and we have finished a 14 day ascending toxicity dose in mice where animals have been given up to three times the therapeutic dose you saw before with absolutely no apparent toxicity at any dose in any organ. So we think in the oncology platform now we`ve achieved three product formulations that have a very exciting future: the vaccine which is demonstrating safety and activity in prostate cancer in a Phase I/II today; the oncolytic virus which will be developed by Cell Genesys up the street from Geron; and our home run, the telomerase inhibitor drugs which should enjoy broad use in combination therapy for virtually all forms of cancer.

Let`s switch now to the second story, the more medium term or mid-term value driver for the company.

Let`s switch now to the second story, the more medium term or mid-term value driver for the company. And the take home message here in regen med, the embryonic stem cell story, is that we`re positioning for clinical development. You know that this is a new paradigm for therapeutics, doing what pharmaceuticals cannot do – literally re-engineer tissue damaged by chronic disease, that is not an achievable objective for pharmacology. The scaleability of embryonic stem cells allows for the first time a high margin, low cost of goods, product based business model which has never been achieved before in cell therapy.

We`ve demonstrated scalable manufacturing methods for seven different differentiated cell types, the therapeutic cells, that we make from our embyronic stem cell lines and four of those seven are now in proof of concept in animal testing. Two of our stem cell lines have been now, are now fully qualified for use in humans, having passed all of the tests FDA asked us to subject them to to demonstrate their safety for human use. We`ve made dramatic progress in one of the cell types, the oligodendrocyte to treat spinal cord injury which will be our first entry into human clinical trials, and, like the oncology platform we were first in this field; we were funding this research since 1995 and because of that we have a controlling intellectual property estate. So let`s look now at what we`ve actually achieved. You know, the promise of this technology just a few years ago was that embryonic stem cells derived from three day old discarded embryos from IVF clinics would be a self-renewing source for the scalable manufacturing of replacement cells for all tissues in the body. Now that was really a tall order. What have we done to validate that progress–promise?

Well, first, we`ve now made in vitro seven different differentiated cell types and without exception, every one of those cell types has spot-on normal human cell biology. We`ve further learned how to scale and produce them and to purify them for animal testing in major diseases with major unmet medical needs, starting for, from liver cells for drug development all the way through diabetes, Parkinson`s Disease, heart failure and so on. And I`m, now, take you through a few of these cell types to give you a feel as to where we are.

First are liver cells. Now you know that drug discovery is hampered by not having an, a source of liver cells to not only test for liver toxicity but more importantly to completely to find hepatic metabolism of drugs before patients are entered into Phase I trials. We have developed such a platform. The stem cell derived hepatocytes make all of the superficial markers that human hepatocytes from biopsy make as well as expressing the full complement of Phase I and Phase II drug metabolizing enzymes. So we`re now seeking a partner, perhaps in the UK, that will help us format these cells for medium to high throughput screening and beta test them in the pharmaceutical sector in the coming months.

We`ve derived hematopoetic cells, blood forming cells. These are in animal studies in Canada where we`re demonstrating engraftment and the production of all three blood lines. The reason for deriving these cells is our approach to avoid immune rejection of the therapeutic cell. We know from bone marrow transplant studies that if I receive a low dose of these hematopoetic cells made from stem cell line A, I will become permanently tolerant, immunologically, to any therapeutic cell made from that same stem cell line. So this is how we plan to avoid the exposure of costly and toxic immune suppressive agents to the patients who`ll be taking the therapeutic cells. In our Geron Biomed subsidiary in the UK, we`ve published this summer on making bone forming cells, osteoblasts. The bone that these cells make is spot-on normal bone of humans as assayed by x-ray defraction. These will be put in animal studies of nonunion fractions-- fractures in the coming months.

We`ve talked for some time about insulin producing islet cells to treat diabetes and I can tell you now that although the efficiency is still low, we have produced insulin secreting and glucagon secreting cells - the cells of the islets in sufficient quantity to move into animal studies and I can also tell you now that these islet cells are glucose responsive. They secret insulin in proportion to the amount of glucose in the media, which is, of course, the gold standard for real human functioning islets. Now in contrast to many of the other diseases, diabetes is one in which we already have human proof of concept, the Edmonton Protocol, in which cataveric islets are injected into the livers of diabetic patients. This will take the place of that approach.

Dopaminergic neurons for Parkinson`s Disease. We have published now on the fact that these cells are completely normal. Dopaminergic neurons, they make dopamine. They have tyrosine hydroxylase. They respond to dopamine, they make synapses. They do all the right things that dopaminergic neurons are supposed to do. We`re at, in now animal proof of concept studies at Geron where we are transplanting these cells to the deep nuclei that are the source of the problem in Parkinson`s Disease and we are getting exuberant outgrowth of human dopaminergic neurons which we hope to demonstrate in more chronic studies will actually re-engineer including to the cortex the entire dopaminergic circuit which is abnormal in Parkinson`s Disease

Cardiomyocytes. Heart muscle cells which spontaneously contract and have, again, completely normal cell biology, including dose response curves to heart drugs clinically used today. These cell types are also in animal studies where we`re showing engraftment of these human cells, and, more importantly, integration of them with the animal`s normal myocardium, electrically and mechanically. The next milestone will be the announcement of the results of these cells transplanted into mouse models of heart failure where we hope to demonstrate improvement in cardiac output, again through specific transplantation of cell types.

Our most advanced cell type is the oligodendrocyte, the cell that makes myelin, or the insulation that surrounds neurons. The objective of the exercise here is to apply these cells to spinal cord injury which we`ve now done in a validated model of human acute spinal cord injury. These animals under anesthesia are given a uniform blow to the spine which results - in red - into a permanent paresis of the animal. And I`ll show you a movie in a moment to demonstrate the best that the control animals get. In contrast - in blue - animals who get the human cells have a statistically significant improvement in their function, which I`ll also illustrate now with a movie that tells this story in a few words.

First, here are the animals that get no treatment. The left lower limb is completely paralyzed, the animal cannot lift its tail off the bottom of the cage. This is permanent. This is the best the animals get. This is at nine weeks after injury. In contrast, animals who receive the oligodendrocytes lift their tail completely off the cage, bear full weight on both lower extremities and although you can`t see this at the very end , they`re actually able to stand on their hind paws, a dramatic result. When we sacrifice the animal and ask why is this happening, we see that the lesion site is actually containing regenerating rat neurons - a very important endpoint - due to the injection of the human cells, and, at the same injury site we now see human myelin surrounding the previously unmyelinated injured animal neurons. We also have data now in a second animal model, the [shiverer] mouse which is genetically unable to make mouse myelin, and we see the same myelination pattern which obviously must come from human cells.

We now have, as I mentioned, two lines, different lines, both derived at Wisconsin, that are qualified for human use, having passed the full battery of tests that rule out human, porcine, bovine or murine viruses. We are making new lines as we speak that have never seen any animal material and as such will be [GNP] in their characteristics. The, in terms of the scaleability, let me give you one example. If we took one master cell bank of embryonic stem cells and devoted all of them at current efficiencies to the production of the glial progenitor cells that we use for spinal cord injury, one master cell bank would produce enough doses of oligodendrocytes to serve as the entire prevalence of spinal cord injury in the United States. So the take home message of this manufacturing capability is that these cells are really the monoclonal antibodies of cell therapy because of the scaleability and low cost production that`s never been possible with cells before. That enables us to segue from the traditional, individualized cell therapy production schema where there are more people per patient making the dose to an automated, closed system that gives us batch production, multi doses of cells per run.

Our patent portfolio is enormous. The numbers of course speak to themselves, but more importantly is the characteristic of the claims - they`re broad, they`re unrestricted, they`re well exemplified by the body of the patent and not only give us a dominant position today, but will give us freedom to operate and will withstand subsequent challenges as these programs move into the clinical and commercial realms

So, in closing, the take home message for oncology, the taproot of Geron based on telomerase is that we`ve clinically validated this target. The vaccine trial at Duke shows safety and efficacy against metastasizing cancer. The oncolytic virus has been demonstrated by our partners to be safe, efficacious, and synergistic with chemotherapy. The inhibitor drugs are a novel class of nontoxic drugs that have potential for broad clinical application because every cancer type obligatorily depends upon telomerase to progress, and these drugs shut it down.

On the regen med side, we are positioning for clinical development. We`re doing the things now that will enable us to move these products, cell type by cell type, through the regulatory pathways which, by the way, are well defined. This is not a black box of how we get cells approved. The rules have been codified in over 15 years of prior cell therapy attempts. This is going to fall right into the realm of biological drugs. So we can make seven different cell types for therapy. They all have normal cell biology, we can make them scalably and we have animal proof of concept for four of them. We have two lines fully qualified for human use. We think the spinal cord application will be first to the clinic which will be a dramatic human proof of concept for us, much like the vaccine is for the cancer vaccine. We have multi dose production lots that for the first time enable a high margin, low cost of goods, product based business model for cell therapy where the output of the production is vials of cells shipped frozen for off the shelf use in medical centers. And, of course, the intellectual property estates are controlling for both programs. We have a balance, cash, a balance sheet and a cash burn that enables us to create a sustainable environment for product development. We`ve done the hard but the right thing in reducing our burn twice, once last year in June and once this year in January, as we shift from discovery to product development. So going forward you will hear more about the vaccine trial as we finish enrolling the Phase I/II patients and can share with you the clinical results in addition to the safety and immunologic responses you`ve heard today. You`ll hear more publications and more talks about 163 and 163L as we move forward in our IND enabling studies with those compounds and hopefully you`ll hear more about Cell Genesys`s plan to develop the oncolytic virus clinically in cancer.

On the regen med side, you`ll hear more publications on animal proof of concept. We hope soon to talk about the impact of cardiomyocytes on cardiac output in animal models of heart failure. The publications for the spinal cord injury model will appear in peer reviewed journals. We hope to be able to describe new cell lines derived in the United States that are [GNP] and that are complete–are made under completely defined conditions. Again, all elements toward the goal of positioning for clinical development.

In another trial not sponsored by Geron at Duke in renal cell carcinoma when the patients were immunized with total tumor RNA we knew that they had responses, responses to laminen and specific antigens in renal cell carcinoma that had been known, what we didn`t know until we did the analysis though was that the dominant antigen in the total tumor RNA in these patients is in fact telomerase - and these patients are enjoying clinical responses.

So those two lines of evidence show that when you target telomerase you get effective anti tumor immunity and no side effects.

Let`s turn quickly to the oncolytic virus. This is a genetically engineered virus in which we control the replication of the virus with the telomerase promoter, the on/off switch. So the only way the virus can replicate and kill the cell it`s replicating in is if it`s in a telomerase positive tumor cell. And we and GTI/Novartis have published on the specificity of this virus before. So what I want to show you here is work also done at GTI that shows synergy of this virus with doxyrubicin. So here we`re measuring tumor volume, the spread or growth of the tumor, of a liver cancer in an animal. The virus alone is shown here in blue with a effect that is significant over control. Here is doxyrubicin by itself. At this dose slightly more potent than the virus by itself. But in these two lines we see the combination of that dose of virus and that dose of doxyrubicin, which is literally complete control of the tumor. Very dramatic synergy. And, again, because we control the replication of the virus elegantly to telomerase, there`s no adverse events in these animals. So we look forward to the continued development of this technology with Cell Genesys

Now the final entry in the oncology space we think are our home run compounds, GRN 163 and its lipidated cousin GRN 163L. Now it`s now agreed upon worldwide by basic scientists and oncologists itself, themselves, that, that cancer progression is inexorably linked to telomerase activity. It is still the only universally validated cancer target. We have worked for a long time to try to generate drugs that are as specific against telomerase as telomerase is specific to cancer to leverage that value of that target. And we have done that with these two compounds which are potent, specific, and absolutely nontoxic inhibitors of telomerase. We have literally demonstrated that 163 is active in vitro against all of the major human cancers in man shown here. We`ve begun now to do animal studies with some of these tumors, and these six cancers are effectively suppressed in vivo in animal models with GRN 163: brain cancer, prostate cancer, lymphoma, myeloma, liver cancer and cervical cancer, and I`ll show you a bit of that data today to give you a feel for how dramatic this compound may be clinically.

This is a picture of 163. A space occupying molecular model. It`s a 13 mer oligonucleotide, 13 building blocks. It is not an antisense molecule. It has no antisense activity whatsoever. It is a direct competitive inhibitor at the active site of telomerase and it is the only molecule in the human body to which this compound binds. It is effective both intratumorally, and I`ll show you that data in a moment, as well as parenterally. We are doing multiple GLP toxicology studies now, we have multiple manufacturing contracts in place and have IP protection not only for the compound but for the unique chemistry that makes it so potent and specific, as well as the target and issued IP for the clinical use of this compound to treat human patients with cancer. Now we`ve shown these data before, the intratumoral application of 163 in animals bearing a very malignant form of brain cancer and the results are quite striking. All the control animals are dead by day 43, 5 of 7 of the treated animals are alive and well at the time they are sacrificed by protocol at which time they are examined histologically for any evidence of residual tumor, and 5 of the 7 animals show zero tumor in their brain. Now that wouldn`t be so surprising if 163 were a toxic compound, but those same animals show absolutely no adverse reactions whatsoever. This is truly a new paradigm in chemotherapy for cancer: specificity, efficacy and the absence of toxicity.


Some new data that I`ll share with you today from Sloan Kettering here in New York in Malcolm Moore`s lab. This is disseminated multiple myeloma done in a way that the colors indicate the spread of the tumor in the animal - so blue color going to red is the presence of the multiple myeloma in the animal`s body. So here on the control animal, as you can see, dramatic spread of the cancer throughout the animal. Here is the maximally toxic--tolerated dose of doxyrubicin, the best compound to be used in this form of cancer and it`s actually somewhat worse than PBS control. In contrast, seven days of treatment with 163 systemically produces this dramatic result, colorometrically that you can see visually , the absence of blue which when quantified shows dramatic efficacy 30 days after the onset of, after the introduction of the cancer. So another example of the systemic potency of 163 as a stand-alone agent in a remarkably malignant tumor which outperforms doxyrubicin at its maximally tolerated dose, and, of course, we enjoy a substantial survival benefit which obviously is the exercise at the end of the game. Now 163L which we used to call 719 is the lipidated version of 163. It`s the same molecule, same chemistry, but we hang covalently a lipid molecule on one end of the drug. This has enormous impact and we are now doing head-to-head studies of 163 vs. 163L in tox, in efficacy, in synergy, in PK, so that we choose the best compound to move forward into the clinic. Clearly the drug has enhanced bioavailability, which I`ll show you, and potency. It also has manufacturing advantages. It`s less expensive to make and, of course, was designed for systemic administration. So here`s the kind of data we`re getting when we compare 163 head-to-head with 163L. Blue is PBS control, the pink is 163 and the green is 163L at a lower dose than 163 done in the same model. So what you see is a low dose of 163L is much more effective than a higher dose of 163 in inhibiting telomerase in the cells in the animal, in the, in the cancer. Also, the length of telomeres, telomere shortening, which is the mechanism of action, at the lower dose of 163 is more potent than the higher dose of – I`m sorry – the lower dose of 163L is more effective at shortening telomeres than the higher dose of 163. If we now look at the whole animal data, we see the same story. The lower dose of 163L is more potent in terms of tumor inhibition in the animal than the higher dose of 163. So those are the kind of data that we`re now getting that augur well for the clinical introduction of the lipidated form of this bio-molecule.


We are now in the toxicity phase of testing 163L and we have finished a 14 day ascending toxicity dose in mice where animals have been given up to three times the therapeutic dose you saw before with absolutely no apparent toxicity at any dose in any organ. So we think in the oncology platform now we`ve achieved three product formulations that have a very exciting future: the vaccine which is demonstrating safety and activity in prostate cancer in a Phase I/II today; the oncolytic virus which will be developed by Cell Genesys up the street from Geron; and our home run, the telomerase inhibitor drugs which should enjoy broad use in combination therapy for virtually all forms of cancer.

Let`s switch now to the second story, the more medium term or mid-term value driver for the company. And the take home message here in regen med, the embryonic stem cell story, is that we`re positioning for clinical development. You know that this is a new paradigm for therapeutics, doing what pharmaceuticals cannot do – literally re-engineer tissue damaged by chronic disease, that is not an achievable objective for pharmacology. The scaleability of embryonic stem cells allows for the first time a high margin, low cost of goods, product based business model which has never been achieved before in cell therapy.

We`ve demonstrated scalable manufacturing methods for seven different differentiated cell types, the therapeutic cells, that we make from our embyronic stem cell lines and four of those seven are now in proof of concept in animal testing. Two of our stem cell lines have been now, are now fully qualified for use in humans, having passed all of the tests FDA asked us to subject them to to demonstrate their safety for human use. We`ve made dramatic progress in one of the cell types, the oligodendrocyte to treat spinal cord injury which will be our first entry into human clinical trials, and, like the oncology platform we were first in this field; we were funding this research since 1995 and because of that we have a controlling intellectual property estate. So let`s look now at what we`ve actually achieved. You know, the promise of this technology just a few years ago was that embryonic stem cells derived from three day old discarded embryos from IVF clinics would be a self-renewing source for the scalable manufacturing of replacement cells for all tissues in the body. Now that was really a tall order. What have we done to validate that progress–promise?

Well, first, we`ve now made in vitro seven different differentiated cell types and without exception, every one of those cell types has spot-on normal human cell biology. We`ve further learned how to scale and produce them and to purify them for animal testing in major diseases with major unmet medical needs, starting for, from liver cells for drug development all the way through diabetes, Parkinson`s Disease, heart failure and so on. And I`m, now, take you through a few of these cell types to give you a feel as to where we are.

First are liver cells. Now you know that drug discovery is hampered by not having an, a source of liver cells to not only test for liver toxicity but more importantly to completely to find hepatic metabolism of drugs before patients are entered into Phase I trials. We have developed such a platform. The stem cell derived hepatocytes make all of the superficial markers that human hepatocytes from biopsy make as well as expressing the full complement of Phase I and Phase II drug metabolizing enzymes. So we`re now seeking a partner, perhaps in the UK, that will help us format these cells for medium to high throughput screening and beta test them in the pharmaceutical sector in the coming months.

We`ve derived hematopoetic cells, blood forming cells. These are in animal studies in Canada where we`re demonstrating engraftment and the production of all three blood lines. The reason for deriving these cells is our approach to avoid immune rejection of the therapeutic cell. We know from bone marrow transplant studies that if I receive a low dose of these hematopoetic cells made from stem cell line A, I will become permanently tolerant, immunologically, to any therapeutic cell made from that same stem cell line. So this is how we plan to avoid the exposure of costly and toxic immune suppressive agents to the patients who`ll be taking the therapeutic cells. In our Geron Biomed subsidiary in the UK, we`ve published this summer on making bone forming cells, osteoblasts. The bone that these cells make is spot-on normal bone of humans as assayed by x-ray defraction. These will be put in animal studies of nonunion fractions-- fractures in the coming months.

We`ve talked for some time about insulin producing islet cells to treat diabetes and I can tell you now that although the efficiency is still low, we have produced insulin secreting and glucagon secreting cells - the cells of the islets in sufficient quantity to move into animal studies and I can also tell you now that these islet cells are glucose responsive. They secret insulin in proportion to the amount of glucose in the media, which is, of course, the gold standard for real human functioning islets. Now in contrast to many of the other diseases, diabetes is one in which we already have human proof of concept, the Edmonton Protocol, in which cataveric islets are injected into the livers of diabetic patients. This will take the place of that approach.

Dopaminergic neurons for Parkinson`s Disease. We have published now on the fact that these cells are completely normal. Dopaminergic neurons, they make dopamine. They have tyrosine hydroxylase. They respond to dopamine, they make synapses. They do all the right things that dopaminergic neurons are supposed to do. We`re at, in now animal proof of concept studies at Geron where we are transplanting these cells to the deep nuclei that are the source of the problem in Parkinson`s Disease and we are getting exuberant outgrowth of human dopaminergic neurons which we hope to demonstrate in more chronic studies will actually re-engineer including to the cortex the entire dopaminergic circuit which is abnormal in Parkinson`s Disease

Cardiomyocytes. Heart muscle cells which spontaneously contract and have, again, completely normal cell biology, including dose response curves to heart drugs clinically used today. These cell types are also in animal studies where we`re showing engraftment of these human cells, and, more importantly, integration of them with the animal`s normal myocardium, electrically and mechanically. The next milestone will be the announcement of the results of these cells transplanted into mouse models of heart failure where we hope to demonstrate improvement in cardiac output, again through specific transplantation of cell types.

Our most advanced cell type is the oligodendrocyte, the cell that makes myelin, or the insulation that surrounds neurons. The objective of the exercise here is to apply these cells to spinal cord injury which we`ve now done in a validated model of human acute spinal cord injury. These animals under anesthesia are given a uniform blow to the spine which results - in red - into a permanent paresis of the animal. And I`ll show you a movie in a moment to demonstrate the best that the control animals get. In contrast - in blue - animals who get the human cells have a statistically significant improvement in their function, which I`ll also illustrate now with a movie that tells this story in a few words.

First, here are the animals that get no treatment. The left lower limb is completely paralyzed, the animal cannot lift its tail off the bottom of the cage. This is permanent. This is the best the animals get. This is at nine weeks after injury. In contrast, animals who receive the oligodendrocytes lift their tail completely off the cage, bear full weight on both lower extremities and although you can`t see this at the very end , they`re actually able to stand on their hind paws, a dramatic result. When we sacrifice the animal and ask why is this happening, we see that the lesion site is actually containing regenerating rat neurons - a very important endpoint - due to the injection of the human cells, and, at the same injury site we now see human myelin surrounding the previously unmyelinated injured animal neurons. We also have data now in a second animal model, the [shiverer] mouse which is genetically unable to make mouse myelin, and we see the same myelination pattern which obviously must come from human cells.

We now have, as I mentioned, two lines, different lines, both derived at Wisconsin, that are qualified for human use, having passed the full battery of tests that rule out human, porcine, bovine or murine viruses. We are making new lines as we speak that have never seen any animal material and as such will be [GNP] in their characteristics. The, in terms of the scaleability, let me give you one example. If we took one master cell bank of embryonic stem cells and devoted all of them at current efficiencies to the production of the glial progenitor cells that we use for spinal cord injury, one master cell bank would produce enough doses of oligodendrocytes to serve as the entire prevalence of spinal cord injury in the United States. So the take home message of this manufacturing capability is that these cells are really the monoclonal antibodies of cell therapy because of the scaleability and low cost production that`s never been possible with cells before. That enables us to segue from the traditional, individualized cell therapy production schema where there are more people per patient making the dose to an automated, closed system that gives us batch production, multi doses of cells per run.

Our patent portfolio is enormous. The numbers of course speak to themselves, but more importantly is the characteristic of the claims - they`re broad, they`re unrestricted, they`re well exemplified by the body of the patent and not only give us a dominant position today, but will give us freedom to operate and will withstand subsequent challenges as these programs move into the clinical and commercial realms

So, in closing, the take home message for oncology, the taproot of Geron based on telomerase is that we`ve clinically validated this target. The vaccine trial at Duke shows safety and efficacy against metastasizing cancer. The oncolytic virus has been demonstrated by our partners to be safe, efficacious, and synergistic with chemotherapy. The inhibitor drugs are a novel class of nontoxic drugs that have potential for broad clinical application because every cancer type obligatorily depends upon telomerase to progress, and these drugs shut it down.

On the regen med side, we are positioning for clinical development. We`re doing the things now that will enable us to move these products, cell type by cell type, through the regulatory pathways which, by the way, are well defined. This is not a black box of how we get cells approved. The rules have been codified in over 15 years of prior cell therapy attempts. This is going to fall right into the realm of biological drugs. So we can make seven different cell types for therapy. They all have normal cell biology, we can make them scalably and we have animal proof of concept for four of them. We have two lines fully qualified for human use. We think the spinal cord application will be first to the clinic which will be a dramatic human proof of concept for us, much like the vaccine is for the cancer vaccine. We have multi dose production lots that for the first time enable a high margin, low cost of goods, product based business model for cell therapy where the output of the production is vials of cells shipped frozen for off the shelf use in medical centers. And, of course, the intellectual property estates are controlling for both programs. We have a balance, cash, a balance sheet and a cash burn that enables us to create a sustainable environment for product development. We`ve done the hard but the right thing in reducing our burn twice, once last year in June and once this year in January, as we shift from discovery to product development. So going forward you will hear more about the vaccine trial as we finish enrolling the Phase I/II patients and can share with you the clinical results in addition to the safety and immunologic responses you`ve heard today. You`ll hear more publications and more talks about 163 and 163L as we move forward in our IND enabling studies with those compounds and hopefully you`ll hear more about Cell Genesys`s plan to develop the oncolytic virus clinically in cancer.

On the regen med side, you`ll hear more publications on animal proof of concept. We hope soon to talk about the impact of cardiomyocytes on cardiac output in animal models of heart failure. The publications for the spinal cord injury model will appear in peer reviewed journals. We hope to be able to describe new cell lines derived in the United States that are [GNP] and that are complete–are made under completely defined conditions. Again, all elements toward the goal of positioning for clinical development.

Plus 55% - Geron übertrifft meine Erwartungen
Geron erklimmt die nächste Stufe der Erfolgsleiter

Grandiose Daten der experimentellen Krebsvakzine sorgten gestern für einen Kurssprung bei Geron. Damit hat Geron meine Erwartungen nicht nur erfüllt, sondern sogar deutlich übertroffen. Ja, sogar ich war sprachlos, als ich gestern am späten Abend einen Blick in mein Depot warf und mir ein Gewinn von 55 Porzent entgegensprang.

Erst am 19. August hatten wir vom Biotech-Experten (www.Biotech-Experte.de) auf die Aktie des Stammzellenspezialisten Geron aufmerksam gemacht und wie sich gestern zeigte, hat sich unsere Intuition wieder einmal bewahrheitet. Wer damals unsere Ausführungen aufmerksam gelesen hat und sich ein paar Aktien von Geron ins Depot gelegt hat, der konnte gestern an nur einem einzigen Tag einen recht ordentlichen Gewinn von 55 Prozent einfahren.

Die starke Nachfrage nach Aktien des führenden Stammzellenforschers kommt allerdings nicht von ungefähr, auch beruht sie nicht, wie bei so manchem anderen Wert, auf den täglichen Zockerspielchen, mit denen sich Börsenspekulanten ihr tägliches Brot verdienen. Nein, Geron hatte wirklich etwas zu bieten am gestrigen Dienstag.

Auf der derzeit in New York statt findenden UBS Global Life Sciences Konferenz ließ Geron’s CEO Dr. Okarma die Bombe platzen, auf deren Explosion ich schon seit geraumer Zeit gewartet habe. Was der CEO der versammelten Investorengemeinde, rund 3.000 an der Zahl, am frühen Nachmittag präsentierte war dann tatsächlich sehr ermutigend. In einer Phase I/II Studie konnte die Prostatakrebsvakzine von Geron ihr festgesetztes Studienziel erreichen. Die experimentelle Vakzine zielt auf ein Enzym das sich Telomerase nennt und in einer Vielzahl von Krebszellen überexprimiert wird. Wird ein Gen für ein bestimmtes Protein überexprimiert, so bedeutet dies nichts anderes, als dass das Protein, in Geron’s Fall eben die Telomerase, in einer zu großen Mengen in den Zellen vorhanden ist.

Bereits in einer früheren Studie hatte Geron gezeigt, dass die Vakzine die Konzentration an im Blut zirkulierenden Krebszellen bis gegen Null reduzieren kann. Die Patienten waren damals mit einer nur geringen Menge der experimentellen Vakzine behandelt worden. In der neuen Studie wurde eine höhere Konzentration verabreicht, um das Sicherheitsprofil zu evaluieren. Die neuen Daten ergaben ein ähnliches, wenn nicht sogar besseres Ergebnis. Die entgültigen Daten der genannten Studie werden bis Ende des Jahres erwartet. Sollte die Geron Aktie demnächst wieder etwas zurückkommen, könnte sich also ein Einstieg durchaus noch auszahlen.

Bei ähnlich guten finalen Daten mit der Vakzine würde sich auch ein längerfristiges Investment in Geron lohnen, da eine Zulassung dann ziemlich wahrscheinlich wäre. Doch nicht nur die Krebsvakzine macht Geron zu einem interessanten Investment, die jüngsten Erfolge des Unternehmens im Bereich Stammzellen dürften den Weg für die so lange erwartete Stammzellentherapie ebnen. Wer bereits mit dem Marktvolumen der Geron-Entwicklung liebäugelt, sollte zudem nicht vergessen, dass die Vakzine das Zeug zum universellen Krebsimpfstoff besitzt. Telomerase wird nämlich in allen bisher untersuchten Krebszellen überexprimiert, in normalen Zellen dagegen kaum. Aus diesem Grunde dürfte auch das Nebenwirkungsprofil eher gering sein.

Im Hinblick auf die bisher veröffentlichten Daten gehe ich auf jeden Fall davon aus, dass die entgültigen Daten der Prostatastudie ähnlich gut ausfallen werden und dem Impfstoffkandidaten den Weg in Phase III frei machen werden.

In den behandelten Patienten des Phase I/II Versuchs konnte die Vakzine den Anteil der T-Zellen, das sind wichtige Bestandteile des Immunsystems, die u.a. auch Krebszellen abwehren, deutlich erhöhen. Eine T-Zellenkonzentration wie sie normalerweise nur bei der Abwehr von Infektionskrankheiten wie Masern oder Mumps auftritt konnte noch niemals mit einer Krebsvakzine erzielt werden. Bei sämtlichen vakzinierten Patienten traten keine ersichtlichen Nebenwirkungen zutage. Spannend dürfte es allerdings erst zum Ende des Jahres werden, denn dann wird Geron auch die klinischen Ansprechraten vorlegen.

Sollten diese eine positive Korrelation mit der T-Zellenkonzentration aufweisen, dann wäre eine baldige Kommerzialisierung des Produktes wohl nicht mehr allzu fern. Nur für die Indikation Prostatakrebs würde dann ein Marktvolumen von mehr als einer Milliarde Dollar winken. Sollte sich die Vakzine allerdings auch bei anderen Krebsarten erfolgreich testen lassen, dann könnte sich das Marktpotenzial noch wesentlich steigern lassen."

Der Onkologie (Krebsbehandlung).  Guten Tag.  Wie unseren folgenden Lautsprecher, Dr. Thomas Okarma, Präsident und CEO vorstellen von Geron.  Dr. Okarma:  Danke, David, und danke alle für heute kommen.  Ich kenne diese won`tüberraschung Sie aber ich auch werde bilden Aussagen vorwärts schauen, also lenken wir Ihre Aufmerksamkeit auf die Gefahrenfaktoren in unserem 10Q.  Wir haben zwei für Sie und I`m heute zusammenzufassende Geschichten, das geht, mit Onkologie, die Krebsplattform zu beginnen, die wirklich unser naher Bezeichnungswerttreiber ist.  Und die Nehmenhauptanzeige für Sie heute im Krebs ist, daß wir glauben, daß we`ve jetzt klinisch das telomeraseziel validierte.  Sie können wissen, daß telomerase heute das einzige bekannte Universalkrebsziel, Bedeutung, die it`s bleibt sehr, die selten in allen Krebszellen und in den normalen Zellen ausgedrückt werden.  Wir haben drei therapeutische Programme, die unterschiedliche Elemente von telomerase im Krebs adressieren.  Das vorgerückteste ist unser Versuch der Phase I/II im Prostatakrebs am Herzog, in dem darstellende und I`ll we`re das jetzt Ihnen die Daten heute geben, dieser telomeraseimpfstoff, Schutzimpfung verursacht sicher immune Antworten des starken Anti- Krebses bei vorgerückten Krebspatienten.  Wir haben ein oncolytic Virusprogramm außerdem, zuerst genehmigt zu GTI/Novartis, jetzt erworben von Cell Genesys, in der wir jetzt in der mehrfachen drastischen Wirksamkeit und in der Synergie der Untersuchungen an Tieren mit Chemotherapie gezeigt haben.  Und zuletzt, sind unser Ausgangsdurchlauf, die telomerasehemmnisdrogen, 163 und die neuere Variante 163L. buchstäblich, diese Mittel in vitro gegen alle menschlichen hauptsächlichkrebse wirkungsvoll und in den Tiermodellen sind in allen sechs Modellen wirkungsvoll gewesen, daß wir bis jetzt diese Drogen herausgestellt haben.  So gehen let`s in dieses ein wenig tiefer und rufen selbstverständlich zurück, daß telomerase die Hahnwurzel von Geron ist und da so wir einen steuernden zu hörenden Zustand des geistigen Eigentums auf der Plattform und dem Produktyou`re ungefähr haben.  Der Versuch am Herzogwe`ve beschrieben im April.  Es ist ein Zweiphasenversuch mit einer niedrigen Dosisgruppe von 12 Patienten und einer Gruppe der hohen Dosis von 12.  Wir berichteten einige über der einleitenden Resultate zurück im April, dem I`ll Sie jetzt von erinnern.  Zuerst alle Patienten in der niedrigen Dosisgruppe - alle 12 von ihnen - reagiert mit den kräftigen immunen Antworten spezifisch zum telomerase.  Alle diese roten Stäbe sind t-Zellen, die spezifisch zum telomerase in allen Patienten reagieren, und diese ist bedeutend, weil diese vorgerückte Krebspatienten sind, die durch Definitionsimmunes unterdrückt sind.  Wir zeigten auch in der ersten niedrigen Dosisgruppe, daß alle 7 der Patienten, die verteilende Niveaus der metastasierenden Prostatakrebszellen erhöht hatten, jene Niveaus hatten, die bis null zusammenfallend mit der Schutzimpfung verringert wurden.  Einige dieser Patienten verloren über tausend Falte der verteilenden Prostatakrebs-Tumorzellen?  kein Patient reagierte nicht auf diesen Impfstoff in so.  Die neuen Daten, die ich mit Ihnen heute teilen kann, sind der Anfang der Resultate von der zweiten Phase, die sogenannte Gruppe der hohen Dosis, die sechs Impfstoffe anstelle von drei in der niedrigen Dosisgruppe empfing.  Und es gibt einige Punkte des hier zu bildenden Wertes.  Zuerst ist das absolute Niveau der t-Zellen, die wir in diesen Themen in der Gruppe der hohen Dosis erzeugen, zwischen einem und zwei Prozent ihrer Gesamtt-Zellenlache;  that`s erzielt nie in der Krebsschutzimpfung.  That`s das Niveau der t-Zellenreaktivität, die Sie mit Masern- oder Mumpsschutzimpfungen sehen.  Zweitens scheint die Dauer der immunen Antwort, sehr langes gelebt zu sein, gut, nachdem die 6. Schutzimpfung und tatsächlich das stabilisierende Niveau der t-Zellenimmunität bei den Patienten der hohen Dosis - bei 13 Wochen - höher als das Höchstniveau ist, das wir in der niedrigen Dosisgruppe erhielten, in der wir die Wirksamkeit hinsichtlich der verteilenden Tumorzellen demonstrierten.  So regte we`re sehr das auf, indem es einfach drei weitere Einspritzungen der gleichen Zahl von Zellenwe`ve gegangen von einer kurzen niedrigen aber wirkungsvollen immunen Antwort der Bezeichnung bis eine hinzufügte, die scheint, lang gelebt zu werden.  So stimmte Aufenthalt später im Jahr als wir jetzt ab, wenn wir die Resultate des Restes der Gruppe und des Fokus der hohen Dosis auf klinischen Antworten vorstellen.  Wir haben gute Stellvertreter jetzt für immune Antwort, die der Hauthautausschlag ist, den, diese Patienten am Einspritzungaufstellungsort sich entwickeln.  Wir haben wirklich diese verzögerte Überempfindlichkeitreaktion und sie biopsiert, um Zellen des telomerase T Anti- zu enthalten gezeigt.  So haben wir jetzt ein Sichtmaß der immunen Antwort.  Und keine der Patienten in der niedrigen Dosisgruppe oder in der Gruppe der hohen Dosis haben eine einzelne nachteilige Reaktion erfahren.  Ein anderes Beweisstück auf der vaccine Seite kam zu uns indirekt.  In einem anderen Versuch, der nicht von Geron am Herzog im Nierenzellenkrebsgeschwür gefördert wird, als die Patienten wurden immunisiert mit Gesamttumor-RNS, die wir wußten, daß sie hatten Antworten, laminen Antworten zu und spezifische Antigene im Nierenzellenkrebsgeschwür, das bekannt, was wir didn`t wissen, bis wir die Analyse taten, waren zwar, daß das dominierende Antigen in der Gesamttumor-RNS bei diesen Patienten tatsächlich telomerase ist - und diese Patienten genießen klinische Antworten.  So zeigen jene zwei Linien des Beweises die, wenn Sie telomerase zielen, das Sie wirkungsvolle Anti- Tumorimmunität und keine Nebenwirkungen erhalten.  Umdrehung Let`s schnell zum oncolytic Virus.  Dieses ist ein genetisch ausgeführtes Virus, in dem wir die Reproduktion des Virus mit dem telomeraseförderer steuern, der AN/CAus-Schalter.  So die einzige Weise, die das Virus wiederholen kann und die Zellenit`s zu töten, die innen wiederholen, ist wenn it`s in einer positiven Tumorzelle des telomerase.  Und wir und GTI/Novartis haben auf der Besonderheit dieses Virus vorher veröffentlicht.  So was ich zeigen möchte, sind Sie hier die Arbeit, die auch an GTI erledigt wird, das Synergie dieses Virus mit doxyrubicin zeigt.  So hier messende Tumorausgabe des we`re, die Verbreitung oder Wachstum des Tumors, eines Leberkrebses in einem Tier.  Das Virus alleine wird hier im Blau mit einem Effekt gezeigt, der bedeutende Übersteuerung ist.  Ist hier doxyrubicin selbst.  An dieser Dosis etwas stärker als das Virus selbst.  Aber in diesen zwei Linien sehen wir die Kombination dieser Dosis des Virus und dieser Dosis von doxyrubicin, das buchstäblich komplette Steuerung des Tumors ist.  Sehr drastische Synergie.  Und wieder weil wir die Reproduktion des Virus elegant zum telomerase steuern, there`s keine nachteiligen Fälle in diesen Tieren.  So wir schauen vorwärts zur anhaltenden Entwicklung dieser Technologie mit WürfelneueBezeichnung des wobei 163L ZellenGenesys Teil 1/b GRN163/163L nicht des ist des für 719 auch, das wundern wo GRN 719 ist jetzt der entscheidende Eintrag im Onkologieraum wir sind unsere Ausgangsdurchlaufmittel denken, GRN 163 geblieben und sein lipidated Vetter GRN 163L. jetzt it`s jetzt weltweit durch grundlegende Wissenschaftler und Onkologen selbst vereinbarte, selbst, die, daß Krebsweiterentwicklung unerbittlich mit telomerasetätigkeit verbunden wird.  Es ist noch das einzige allgemeinhin validierte Krebsziel.  Wir haben für eine lange Zeit gearbeitet, um zu versuchen, Drogen zu erzeugen, die gegen telomerase so spezifisch sind, wie telomerase zum Krebs zur Hebelkraft spezifisch ist, die Wert dieses Ziels.  Und wir haben das mit diesen zwei Mitteln getan, die starke, spezifische und absolut ungiftige Hemmnisse von telomerase sind.  Wir haben buchstäblich gezeigt, daß 163 in vitro gegen alle menschlichen hauptsächlichkrebse im Mann aktiv ist, der hier gezeigt wird.  We`ve, das jetzt angefangen werden, um Untersuchungen an Tieren mit einigen dieser Tumoren zu tun, und diese sechs Krebse werden effektiv in vivo in den Tiermodellen mit GRN 163 unterdrückt:  Gehirnkrebs, Prostatakrebs, Lymphom, myeloma, Leberkrebs und zervikaler Krebs und Erscheinen I`ll Sie ein wenig diese Daten heute zum Geben Ihnen eines Gefühls für, wie drastisch dieses Mittel klinisch sein kann.  Dieses ist eine Abbildung von 163.  Ein Raum, der molekulares Modell besetzt.  It`s ein Oligonucleotide mit 13 mer, 13 Bausteine.  Es ist nicht ein antisensemolekül.  Es hat keine antisensetätigkeit whatsoever.  Es ist ein direktes konkurrierendes Hemmnis am aktiven Aufstellungsort von telomerase und es ist das einzige Molekül im menschlichen Körper, an den dieses Mittel bindet.  Es ist beide intratumorally und Erscheinen I`ll Sie diese Daten in einem Moment, sowie parenteral wirkungsvoll.  Wir tun mehrfache GLP-Toxikologiestudien jetzt, haben wir mehrfache Herstellungsverträge im Platz und haben IPSCHUTZ nicht nur für das Mittel aber für die einzigartige Chemie, die Marken es so stark und spezifisch, sowie das Ziel und das herausgegebene IP für den klinischen Gebrauch dieses Mittels, menschliche Patienten mit Krebs zu behandeln.  Das jetzt we`ve, das diese Daten vor, die intratumoral Anwendung von 163 in den Tieren tragen eine sehr bösartige Form des Gehirnkrebses und der Resultate gezeigt wird, sind ziemlich auffallend.  Alle Steuertiere sind- bis Tag 43 tot, sind 5 von 7 der behandelten Tiere lebendig und wohl, zu der Zeit als sie durch Protokoll zu welcher Zeit geopfert werden, werden sie histologisch für jeden möglichen Beweis des Resttumors überprüft, und 5 der 7 Tiere zeigen nulltumor in ihrem Gehirn.  Nun da wouldn`t so überrascht, wenn 163 ein giftiges Mittel waren, aber jenes, zeigen die gleichen Tiere absolut keinen nachteiligen Reaktionen whatsoever.  Dieses ist wirklich ein neues Paradigma in der Chemotherapie für Krebs:  Besonderheit, Wirksamkeit und das Fehlen Giftigkeit.  Etwas neue Daten dieser Anteil I`ll mit Ihnen heute von Sloan Kettering hier in neuem York im Labor Malcolm Moore`s.  Dieses ist das verbrittene mehrfache myeloma, das in einer Weise getan wird, daß die Farben die Verbreitung des Tumors im Tier anzeigen - also ist die blaue Farbe, die zum Rot geht, das Vorhandensein des mehrfachen myeloma im animal`skörper.  So hier auf dem Steuertier, wie Sie sehen können, drastische Verbreitung des Krebses während des Tieres.  Ist hier das maximal giftige -- zugelassene Dosis von doxyrubicin, von besten in dieser Form des Krebses zu verwendenden Mittel und von it`s wirklich ein wenig schlechter als PBS-Steuerung.  Demgegenüber produziert sieben Tage der Behandlung mit 163 körperlich dieses drastische Resultat, colorometrically, daß Sie sichtlich sehen können, das Fehlen Blau, deren wenn quantitativ bestimmte drastische Wirksamkeit der Erscheinen 30 Tage nach Angriff, nach der Einleitung des Krebses.  So ein anderes Beispiel der Körperkraft von 163 als alleinstehendem Mittel in einem bemerkenswert bösartigen Tumor, der doxyrubicin an seiner maximal zugelassenen Dosis an Leistung übertrifft und selbstverständlich wir genießen einen erheblichen Überlebensnutzen, der offensichtlich die Übung am Ende des Spiels ist.  Jetzt ist 163L, das wir pflegten, zu nennen 719, lipidated Version von 163.  It`s das gleiche Molekül, die gleiche Chemie, aber wir hängen kovalent ein Lipidmolekül an einem Ende der Droge.  Dieses hat enorme Auswirkung und wir tun jetzt head-to-head Studien von 163 gegen.  163L im tox, in der Wirksamkeit, in der Synergie, in PK, damit wir das beste Mittel beschließen, um vorwärts in die Klinik umzuziehen.  Offenbar hat die Droge Lebenskraft erhöht, die Erscheinen I`ll Sie und Kraft.  Sie hat auch Herstellungsvorteile.  It`s, das weniger kostspielig ist zu bilden und selbstverständlich war für Körperleitung bestimmt.  So here`s die Art von Datenwe`re erhalten, wenn wir 163 head-to-head mit Blau 163L. sind PBS-Steuerung vergleichen, ist der Pink 163 und das Grün ist 163L an einer untereren Dosis als 163 getan im gleichen Modell.  So was Sie sehen, ist eine niedrige Dosis von 163L ist viel wirkungsvoller als eine höhere Dosis von 163 in inhibierendem telomerase in den Zellen im Tier, in, im Krebs.  Auch die Länge von telomeres, telomereshortening, der die Einheit der Tätigkeit ist, an der untereren Dosis von 163 ist stärker als die höhere Dosis von?  I`m traurig?  die unterere Dosis von 163L ist an den Shorteningtelomeres als die höhere Dosis von 163 wirkungsvoller.  Wenn wir jetzt die vollständigen Tierdaten betrachten, sehen wir die gleiche Geschichte.  Die unterere Dosis von 163L ist in Tumorhemmung im Tier ausgedrückt als die höhere Dosis von 163 stärker.  So sind die die Art von Daten, die das jetzt erhaltene we`re, daß augur gut für die klinische Einleitung von Form dieses Biomoleküls lipidated.  Wir sind jetzt in der Giftigkeitphase der Prüfung von 163L und wir haben eine 14 Tagessteigende Giftigkeitdosis in den Mäusen beendet, in denen Tiere bis dreimal die therapeutische Dosis Sie Säge vorher mit absolut keiner offensichtlichen Giftigkeit an jeder möglicher Dosis in irgendeinem Organ gegeben worden sind.  So denken wir im we`ve der Onkologieplattform jetzt erzielten drei Produktformulierungen, die eine sehr aufregende Zukunft haben:  der Impfstoff, der Sicherheit und Tätigkeit im Prostatakrebs in einer Phase I/II heute zeigt;  das oncolytic Virus, das von Cell Genesys herauf die Straße von Geron entwickelt wird;  und unser Ausgangsdurchlauf, die telomerasehemmnisdrogen, die ausgedehnten Gebrauch in der Kombinationstherapie für praktisch alle Formen des Krebses genießen sollten.  Schalter Let`s jetzt zur zweiten Geschichte, zur mittlereren Bezeichnung oder zum Halbzeitwerttreiber für die Firma.  Schalter Teil 2 Stammzellen Let`s jetzt zur zweiten Geschichte, zur mittlereren Bezeichnung oder zum Halbzeitwerttreiber für die Firma.  Und die Nehmenhauptanzeige hier im regen med, die embryonale Stammzellegeschichte, ist dieses we`re, das für klinische Entwicklung in Position bringt.  Sie wissen, daß dieses ein neues Paradigma für Therapeutik ist und tut, was pharmazeutische Produkte nicht tun können?  buchstäblich Re-Ingenieur, den Gewebe durch chronische Krankheit, die beschädigte, ist nicht eine erreichbare Zielsetzung für Pharmakologie.  Das scaleability der embryonalen Stammzellen erlaubt zum ersten Mal einen hohen Seitenrand, niedrige Kosten Waren, Produkt gegründetes Geschäftsmodell, das nie vorher in der Zellentherapie erzielt worden ist.  We`ve zeigte scalable Produktionsmethoden für sieben unterschiedliche unterschiedene Zellenarten, die therapeutischen Zellen, die wir von unseren embyronic Stammzellelinien bilden und vier von jenen sieben jetzt im Beweis des Konzeptes in der Tierprüfung sind.  Zwei unserer Stammzellelinien sind jetzt, werden jetzt qualifiziert völlig für Gebrauch in den Menschen gewesen, nachdem siegeführt die ganze Test-FDA siegeführt hatten, bat uns, sie zu unterwerfen, um ihre Sicherheit für menschlichen Gebrauch zu demonstrieren.  We`ve bildete drastischen Fortschritt in einer der Zellenarten, das oligodendrocyte zur Rückenmarkverletzung der Festlichkeit, die unsere erste Eintragung in menschliche klinische Versuche ist, und, wie der Onkologieplattform, die wir erste auf diesem Gebiet waren;  wir finanzierten diese Forschung seit 1995 und wegen dessen haben wir einen steuernden Zustand des geistigen Eigentums.  So betrachten let`s jetzt, was we`ve wirklich erzielte.  Vor Sie wissen, war die Versprechung dieser Technologie gerade einigen Jahren, daß die embryonalen Stammzellen, die von drei Tagesalten weggeworfenen Embryos von den IVF-Kliniken abgeleitet wurden, eine selbsterneuernde Quelle für die scalable Herstellung der Wiedereinbauzellen für alle Gewebe im Körper sein würden.  Nun da wirklich ein hohes war, bestellen Sie.  Was haben wir getan, um dieses progress?promise zu validieren?  Gut hat erstes, das we`ve, das jetzt in vitro sieben unterschiedliche unterschiedene Zellenarten und ohne Ausnahme, jede jener Zellenarten gebildet wird, Punkt-auf normaler menschlicher Zellenbiologie.  Weiteres gelehrtes We`ve, wie man sie für das Tier einstuft und produziert es und reinigt, das, in den Hauptkrankheiten mit medizinischen Notwendigkeiten des Hauptunmet prüft und an, von den Leberzellen an der Drogeentwicklung vollständig durch Diabetes, Krankheit Parkinson`s, Herzausfall und so weiter abfährt.  Und I`m nehmen Sie jetzt durch einige dieser Zellenarten, um zu geben Ihnen ein Gefühl hinsichtlich, dem wir sind.  Sind zuerst Leberzellen.  Jetzt wissen Sie, daß Drogeentdeckung wird gehemmt indem man nicht hat, eine Quelle der Leberzellen, um auf Lebergiftigkeit aber zu vollständig nicht nur zu prüfen hepatischen Metabolismus der Drogen wichtiger zu finden, bevor Patienten in Versuche der Phase I angemeldet werden.  Wir haben solch eine Plattform entwickelt.  Die abgeleiteten hepatocytes der Stammzelle bilden alle von den oberflächlichen Markierungen, die menschliche hepatocytes von der Biopsie bilden, sowie, die volle Ergänzung der Droge der Phase I und der Phase II ausdrückend, die Enzyme umwandelt.  So we`re, das jetzt einen Partner, möglicherweise in Großbritannien sucht, das uns hilft, diese Zellen für Mittel zur hohen Durchsatzsiebung und zum Betatest zu formatieren sie im pharmazeutischen Sektor in den kommenden Monaten.  We`ve leitete hematopoetic Zellen, das Blut ab, das Zellen bildet.  Diese in den Untersuchungen an Tieren in Kanada in dem das we`re sind, das engraftment und die Produktion aller drei Blutlinien zeigt.  Der Grund für das Ableiten dieser Zellen ist unsere Annäherung zum Vermeiden der immunen Ablehnung der therapeutischen Zelle.  Wir wissen von den Knochenmarktransplantstudien, daß, wenn ich eine niedrige Dosis dieser hematopoetic Zellen empfange, die von Stammzellelinie A gebildet werden, ich immunologisch dauerhaft tolerant werde zu jeder therapeutischen Zelle, die von dieser gleichen Stammzellelinie gebildet wird.  So ist dieses, wie wir planen, die Belichtung von teurem zu vermeiden und giftige immune unterdrückende Mittel zum Patientenwho`ll die therapeutischen Zellen nehmen.  In unserer Tochtergesellschaft Geron Biomed in Großbritannien, veröffentlichte we`ve diesen Sommer auf dem Bilden des Knochens, der Zellen, osteoblasts bildet.  Der Knochen, den diese Zellen bilden, ist Punkt-auf normalem Knochen der Menschen, wie durch Röntgenstrahldefraction geprüft.  Diese werden in Untersuchungen an Tieren der nicht organisierten Brüche -- Brüche in den kommenden Monaten eingesetzt.  We`ve sprach während einiger Zeit über Insulin, Inselzellen produzierend mit Festlichkeitdiabetes und ich kann Sie, nun da, obgleich die Leistungsfähigkeit noch niedrig ist, wir Insulin absondernd und produziert haben Glucagonabsondernzellen erklären - die Zellen der kleinen Inseln in der genügenden Quantität zum Bewegen in Untersuchungen an Tieren und in mich können Ihnen auch erklären, nun da diese Inselzellen die entgegenkommende Glukose sind.  Sie geheimes Insulin im Verhaeltnis zu der Menge der Glukose in den Mitteln, die ist, selbstverständlich die Goldaktie für reale menschliche arbeitende kleine Inseln.  Jetzt im Gegensatz zu vielen der anderen Krankheiten, ist Diabetes einer, in dem wir bereits menschlichen Beweis des Konzeptes haben, das Edmontonprotokoll, in dem cataveric kleine Inseln in die Lebern der zuckerkranken Patienten eingespritzt werden.  Dieses findet dieser Annäherung statt.  Dopaminergische Neuronen für Krankheit Parkinson`s.  Wir haben jetzt auf der Tatsache veröffentlicht, daß diese Zellen vollständig normal sind.  Dopaminergische Neuronen, bilden sie Dopamin.  Sie haben Tyrosinhydroxylase.  Sie reagieren auf Dopamin, sie bilden Synapsen.  Sie tun alle rechten Sachen, die dopaminergische Neuronen tun sollen.  We`re an, im jetzt Tierbeweis des Konzeptes studiert bei Geron, in dem wir diese Zellen zu den tiefen Kernen verpflanzen, die die Quelle des Problems in der Krankheit Parkinson`s sind und wir erhalten exuberant Ergebnis der menschlichen dopaminergischen Neuronen, die wir hoffen, in den chronischeren Studien zu demonstrieren werden wirklich der Re-Ingenieur, der zur Rinde den gesamten dopaminergischen Stromkreis umfaßt, der in der Krankheit Cardiomyocytes Parkinson`s anormal ist.  Herzmuskelzellen, die spontan wieder Vertrag vollständig normalen Zellenbiologie abschließen und haben, einschließlich des Ansprechens auf die Dosis kurvt zu den Herzdrogen, die klinisch heute benutzt werden.  Diese Zellenarten auch in den Untersuchungen an Tieren in denen das we`re sind, das wichtiger engraftment dieser menschlichen Zellen und, der Integration von ihnen mit dem normalen Myocardium der animal`s, elektrisch und mechanisch zeigt.  Der folgende Meilenstein ist die Ansage der Resultate dieser Zellen, die in Maus verpflanzt werden, die, Modelle des Herzausfalls, in dem wir hoffen, Verbesserung in der Herzleistung zu demonstrieren, wieder durch spezifische Versetzung der Zelle schreibt.  Unsere vorgerückteste Zellenart ist das oligodendrocyte, die Zelle, die myelin bildet, oder die Isolierung, die Neuronen umgibt.  Die Zielsetzung der Übung hier ist, diese Zellen an der Rückenmarkverletzung anzuwenden, die das we`ve jetzt erfolgt in einem validierten Modell der menschlichen akuten Rückenmarkverletzung.  Diese Tiere unter Anästhesie werden einen konstanten Schlag
