Emergency Use Authorization Granted For BioCryst's Peramivir

BIRMINGHAM, Ala., Oct 23, 2009 /PRNewswire-FirstCall via COMTEX/ ----BioCryst Pharmaceuticals, Inc. (Nasdaq: BCRX) today announced that the U.S. Food and Drug Administration (FDA), in response to a request from the U.S. Centers for Disease Control and Prevention (CDC), has issued an emergency use authorization (EUA) for the investigational anti-viral drug intravenous (i.v.) peramivir in certain adult and pediatric patients with confirmed or suspected 2009 H1N1 influenza infection who are admitted to a hospital.

Specifically, i.v. peramivir is authorized only for hospitalized adult and pediatric patients for whom therapy with an i.v. drug is clinically appropriate, based on one or more of the following reasons:

  1. the patient is not responding to either oral or inhaled anti-viral
  therapy, or
  2. when drug delivery by a route other than an intravenous route -- e.g.,
  enteral (absorbed by the intestines) or inhaled -- is not expected to be
  dependable or feasible;
 
  3. for adults only, when the clinician judges i.v. therapy is appropriate
  due to other circumstances.

In advance of any U.S. Government order that may come from the ongoing request for proposal (RFP) negotiations, BioCryst has donated and transferred to the Department of Health and Human Services (HHS) an initial supply sufficient for 1,200 courses of i.v. peramivir 600 mg once-daily for five days. This transfer was made under the development contract with HHS to allow doctors and patients near-term access to the drug, and is separate from the RFP process.

"The issuance of this EUA is important because it makes peramivir a treatment option for physicians in the U.S. during the ongoing influenza health emergency," said Jon P. To prepare for peramivir orders that BioCryst may receive from the U.S. or other governments during this pandemic emergency, BioCryst is completing production of approximately 130,000 courses of i.v. peramivir and is prepared to make more, if required.

FDA Approves Seasonal Influenza Vaccine Fluarix for Pediatric Use

SILVER SPRING, Md., Oct 19, 2009 /PRNewswire-USNewswire via COMTEX/ ----The U.S. Food and Drug Administration today approved use of the seasonal influenza vaccine Fluarix for children ages 3 years to 17 years. Previously, this vaccine, which contains inactivated (killed) influenza A and B viruses, had been approved for use in adults, ages 18 years and older.

The safety and effectiveness of Fluarix for use in children ages 3 years and older is documented by a U.S. study comparing 2,115 children who received Fluarix with 1,210 children who received Fluzone, a different influenza vaccine already licensed by the FDA for use in children ages 6 months and older. Study results showed that children 3 years and older vaccinated with Fluarix and Fluzone produced similar amounts of antibodies in the blood at levels considered likely to be protective against seasonal influenza.

Fluarix is a seasonal influenza vaccine not intended to protect against the 2009 H1N1 influenza virus.

"This approval of an additional seasonal influenza vaccine for children provides help in protecting them against influenza," said Karen Midthun, M.D., acting director of the FDA's Center for Biologics Evaluation and Research. "Children are very vulnerable to the influenza virus and are more likely to be hospitalized for associated problems."

With today's approval, there are now four companies approved by the FDA to manufacture seasonal influenza vaccine for use in children.

Influenza is far more dangerous than the common cold for children, who often require medical care, especially if they are younger than 5 years. It is best to vaccinate children each fall, but vaccination also can occur in the winter months when influenza season often peaks.

Common adverse events experienced after administration of Fluarix are typical of those for flu shots and include pain, redness, and swelling at the injection site as well as irritability, loss of appetite, and drowsiness.

Because Fluarix contains a small amount of egg protein, it should not be administered to anyone allergic to eggs or egg products.

Fluarix is manufactured by GlaxoSmithKline Biologicals of Dresden, Germany.

GAITHERSBURG, Md., Oct 22, 2009 /PRNewswire via COMTEX/ ----MedImmune announced today that interim data from human studies of its nasal spray vaccine for the 2009 novel Influenza A (H1N1) virus demonstrate a similar clinical profile in children and adults 2 to 49 years of age as previously studied seasonal formulations of the vaccine. The live attenuated influenza vaccine (LAIV) for the 2009 novel Influenza A (H1N1) virus received approval from the U.S. Food and Drug Administration (FDA) on September 15, 2009.

"MedImmune is pleased to report these findings that demonstrate that the safety profile and immunogenicity of our nasal spray vaccine for the 2009 H1N1 virus is, as expected, similar to our seasonal influenza vaccine," said Filip Dubovsky, M.D., vice president, clinical development, vaccines. "It is rewarding for all of us at MedImmune to see the vaccine being used to help protect the U.S. population from this novel strain of influenza."

Two randomized, double-blind, placebo-controlled trials are ongoing in children (2-17 years) and adults (18-49 years) to evaluate the safety, tolerability, and immunogenicity of vaccine. A total of 326 children and 300 adults were randomly assigned to receive 2 doses of 2009 H1N1 LAIV or placebo 28 days apart. Both clinical trials are collecting data following a second dose of the vaccine.

LAIV for 2009 H1N1 is made using the same process as LAIV for seasonal influenza. The safety of the 2009 H1N1 vaccine was evaluated in the same way that the safety of new seasonal LAIV strains is assessed every year. The H1N1 vaccine was also evaluated in children to further establish its safety profile.

In children, the most common side effects following the first dose of vaccine included headache, runny/stuffy nose, and cough. In adults, the most common side effects were headache, runny nose and decreased activity. Symptoms after the second dose were similar but generally occurred at lower rates in both children and adults. No vaccine-related serious adverse events have been reported in children or adults. The local and systemic symptoms observed are consistent with intranasal vaccine virus replication and are similar to those observed with seasonal LAIV. Serum antibody responses to the vaccine in addition to pre-clinical characterization data are also consistent with previously studied formulations of seasonal LAIV, which has been shown in multiple studies to be safe and effective in eligible children and adults 2 to 49 years of age.

LAIV initiates the immune response in the nose, where the virus enters the body and replicates, and is the key to establishing a vaccine response that will help prevent disease if the person later encounters a circulating H1N1 virus. In response to the vaccine, the body develops mucosal immunoglobulin A (IgA), serum immunoglobulin G (IgG), and cellular immunity to help protect the individual from infection from an influenza virus.

Study results have been submitted to and reviewed by authorities at the FDA, the U.S. Centers for Disease Control and Prevention (CDC), the National Institutes of Health (NIH), and the U.S. Department of Health and Human Services (HHS).

LAIV is different from the injectable influenza vaccine ("flu shot") in that it contains live vaccine virus strains that are specifically designed to prompt the body to begin mounting a protective response, but weakened so as not to cause the flu. It is delivered into the nose, where the influenza virus usually enters the body, rather than by injection. MedImmune's seasonal influenza vaccine is currently licensed in the United States, South Korea, and Hong Kong and MedImmune's H1N1 influenza vaccine is currently licensed in the United States; both are approved for eligible individuals 2 through 49 years of age.

Novartis übernimmt für 125 Mio. USD Mehrheitsbeteiligung an der chinesischen H1N1-Impfstoff-Tochtergesellschaft von Hard to Treat Diseases (HTDS.PK)

Shenzhen, China, November 10 (ots/PRNewswire) - Hard To Treat Diseases (HTDS.PK), http://www.htdsmedical.com, gab heute bekannt, dass Novartis, ein 41 Mrd. USD schweres, internationales Pharmaunternehmen, für 125 Mio. USD einen Anteil in Höhe von 85% an HTDSs H1N1-Impfstoffpartner Zhejiang Tianyuan Bio-Pharmaceutical Co., Ltd., übernommen hat. Das Schweizer Unternehmen Novartis startet eine auf fünf Jahre und 1 Mrd. USD ausgelegte Initiative in China, die zwei Ziele verfolgt: eine drastische Erhöhung der Investitionen in die chinesische F&E-Anlage des Unternehmens und die Absicht, China zu einem der drei weltweiten Spitzenmärkte des Unternehmens zu entwickeln. Novartis berichtete, das Unternehmen habe den Kauf der Mehrheitsbeteiligung an Zhejiang Tianyuan über die vorgeschlagene Übernahme einer Anteilsmehrheit in Höhe von 125 Mio. USD unternommen, um die Präsenz des Unternehmens im Bereich humaner Impfstoffe in China auszubauen. Zhejiang Tianyuan ist ein in privater Hand befindliches Impfstoffunternehmen mit einem wettbewerbsfähigen Produktportfolio und einer Entwicklungspipeline in China, wo Novartis im Bereich Impfstoffe nur begrenzt präsent ist. Tianyuan und Novartis werden beim Aufbau einer Impfstoffindustrie für das in diesem Bereich bisher medizinisch unterversorgte China, dem drittgrössten Impfstoffmarkt, zusammenarbeiten. Die Übernahme des 85-prozentigen Anteils an Tianyuan für einen Betrag von ca. 125 Mio. USD macht eine aufsichtsrechtliche und staatliche Genehmigung in China notwendig.

Tianyuan ist ein in privater Hand befindliches Impfstoffunternehmen, das in China eine Reihe bereits auf dem Markt befindliche Impfstoffprodukte anbietet und F&E-Projekte mit Schwerpunkt auf mehrere vermeidbare virale und bakterielle Erkrankungen durchführt. Experts Say Swine Flu Mutations Do Not Warrant New Alarm

The World Health Organization tried this week to dampen fears about mutations seen in the swine flu virus in several countries, noting that both mutations had been found in very few people.

A change that created Tamiflu resistance has been found in about 75 people around the world, said Dr. Keiji Fukuda, chief flu adviser to the W.H.O.’s director general. Two clusters, in cancer units at Duke University Medical Center in North Carolina and a hospital in Wales, were both among patients whose immune systems had been severely suppressed by cancer treatment; some had had their bone marrow, which produces infection-fighting white blood cells, wiped out so that replacement blood stem cells could be injected.

Such patients are more likely to develop resistant viruses when on Tamiflu because they can not clear a virus on their own. But the mutant strain appears not to spread easily in people with normal immunity, like hospital workers.

“We don’t know the full answer, but it is more likely that we are not seeing a major shift,” Dr. Fukuda said.

Widespread Tamiflu resistance is a serious problem in the seasonal H1N1 virus, but it has not crossed over into the swine H1N1.

Dr. Fukuda also said W.H.O. scientists were “not sure” of the level of threat posed by a separate mutation that helps the virus reach the lungs. It has been found in Norway, Ukraine, Brazil, China, Japan, Mexico and the United States, in both serious and mild cases.

Experts still need to see whether the mutation — whose shorthand name in virology is D222G or D225G — is becoming more common, and how often it leads to severe disease, he said.

One isolate from Ukraine with the mutation had changed so that swine flu vaccine probably would not protect against it well, Britain’s national medical laboratory reported Friday.

Flus mutate so fast, Dr. Fukuda cautioned, that announcing each change is “like reporting changes in the weather.”

More than 100 million swine flu shots have been administered in 40 countries, and the side effects are similar to or lower than those from regular flu shots, he added.

Six cases of anaphylaxis were widely reported in Canada when it was found that all were from one batch of vaccine, which was recalled. Anaphylaxis is an allergic reaction, usually to egg protein, that can range from mild hives to fatal airway swelling. All six patients recovered, and Dr. Fukuda pointed out that there were 172,000 doses in the batch, most of which caused no problems, so it was unclear whether the cause was a bad batch or just a coincidence.

The stock has been under relentless attack by short sellers and can be bought for about four times earnings. I think after a few more countries order Peramivir and the royalties start to flow from Japan, a short squeeze can carry the stock to $30 over the next 12 months.
