Eine Spekulation auf Umsätze mit der Vogelgrippe stellt die Aktie von Biocryst Pharmaceuticals dar. Das US-Unternehmen verfügt über das Grippemedikament Peramivir, das derzeit auf seine Wirksamkeit gegen die Vogelgrippe getestet wird. Im Januar hat die Firma von der US-Arzneimittelbehörde FDA den beschleunigten Fast-Track-Status für die klinischen Versuche erhalten. Doch Vorsicht: 2002 ist Biocryst mit demselben Medikament in den klinischen Studien der Phase III durchgefallen. Dabei hatte sich herausgestellt, daß durch die Einnahme der Tabletten nicht genug Wirkstoff in den Patienten gelangte. Entwicklungspartner Johnson & Johnson kündigte daraufhin die Zusammenarbeit mit Biocryst, die Firma stellte die Entwicklung ein, die Aktie stürzte ab. Mittlerweile bietet Biocryst Peramivir als Injektion an. Dadurch soll gewährleistet werden, daß genug Wirkstoff in den Körper gelangt. Das scheint auch die Phantasie der Aktionäre anzuregen. Seit den Tiefstständen bei drei Euro im Mai vergangenen Jahres hat die Aktie ihren Wert versechsfacht.

das virus dürfte sich weiterverbreiten und hat schon jetzt nach wenigen Tagen deutlich mehr Menschen infiziert und getötet als alle Vögelinfluenza viren der letzten 10 Jahre zusammen.

hier bleibt man - bis sich entweder eine grosse Verbreitung des Virus entwickelt hat oder bis es den Anschein hat, dass man den Ausbruch (hoffentlich doch noch) unter Kontrolle bekommt.

rein raus ist hier mM der falsche Ansatz; der Wirkstoff des Unternehmens ist gut, die Ergebnisse der Phase II klinischen Prüfung ebenfalls.

Data from Phase 2 Study of Peramivir in Patients Hospitalized with Influenza Presented at the XI International Symposium on Respiratory Viral Infections

BIRMINGHAM, Ala., Feb 23, 2009 /PRNewswire-FirstCall via COMTEX News Network/ -- BioCryst Pharmaceuticals (Nasdaq: BCRX) today announced that the full data set from an exploratory Phase 2 study of peramivir in patients hospitalized for influenza was presented by the study's Principal Investigator, Dr. Michael G. Ison, Assistant Professor, Divisions of Infectious Diseases and Organ Transplantation at Northwestern University Feinberg School of Medicine, during the XI International Symposium on Respiratory Viral Infections taking place in Bangkok, Thailand, February 19 through February 22, 2009. This exploratory Phase 2 trial compared the efficacy and safety of five days of therapy with either 200 mg intravenous (i.v.) peramivir per day, 400 mg i.v. peramivir per day or 75 mg oral oseltamivir twice-a-day, in patients who required hospitalization related to influenza.

The primary objective of the study was to evaluate time to clinical stability, which is a composite endpoint comprised of normalization of temperature, oxygen saturation, respiratory rate, systolic blood pressure and heart rate. This type of endpoint has previously been used in pneumonia studies, but not in influenza. Secondary objectives of the study included evaluation of viral shedding, mortality, clinical relapse and time to resumption of usual activities. The primary efficacy population was defined as patients with confirmed influenza.

As reported in October 2008, there were no statistically significant differences in any of the efficacy endpoints between the three treatment arms, and peramivir was generally safe and well-tolerated at these dose levels. Evaluation of time to clinical stability, the primary endpoint, showed a median of 23.7 hours for peramivir 200 mg, 37.0 hours for peramivir 400 mg and 28.1 hours for oseltamivir (p=0.306). This exploratory endpoint was driven by resolution of fever. Viral shedding (time weighted change from baseline in viral titer) was reduced by a median of -2.0 logs for peramivir 200 mg, -2.1 logs for peramivir 400 mg, and -1.9 logs for oseltamivir (p=0.908). There was no mortality in the primary efficacy population, and there were no clinical relapses. Patients were discharged from hospital after a median of 4.0 days for peramivir 200 mg, 3.8 days for peramivir 400 mg, and 4.0 days for oseltamivir (p=0.994). The median number of days required for resumption of usual activities was 8.8 days for peramivir 200 mg, 9.0 days for peramivir 400 mg, and 13.7 days for oseltamivir (p=0.276).

"We are encouraged by these data and are currently working with the U.S. Food and Drug Administration to determine the next steps for peramivir as a treatment for patients hospitalized with influenza," said Dr. William P. Sheridan, BioCryst's Chief Medical Officer. "There are currently no approved treatment options for the 200,000 patients hospitalized for influenza each year and we believe peramivir may be a potential new option for these patients."

"Peramivir holds promise as a once-daily intravenous treatment that is safe and potentially effective for use in patients with acute influenza, including the elderly and patients with underlying cardiac or pulmonary disease."

The multicenter, randomized, double-blind, double-dummy, active-controlled, Phase 2 study enrolled 137 patients, who tested positive by rapid antigen test (RAT) for influenza and had one or more criteria for hospitalization, namely: age greater than or equal to 60 years, chronic lung disease, congestive heart failure, diabetes mellitus, low oxygen saturation, low blood pressure, or severity of illness requiring supportive care. Of the 137 patients randomized, 122 age 19 to 101 years had influenza confirmed by polymerase chain reaction (PCR) testing and were included in the intent-to-treat infected (ITTI) patient population; 41 patients received oseltamivir 75 mg orally twice-daily, 41 patients received 200 mg i.v. peramivir once-daily and 40 patients received 400 mg i.v. peramivir once-daily. The study was conducted in the United States, Canada, Hong Kong, Singapore, Australia, New Zealand, and South Africa, in 2007 and 2008.

BioCryst plans to present details from the study at the annual meeting of the American Society of Clinical Oncology, which will be held from May 29 to June 2 in Orlando, Fla.

als Folge der (erwarteten) starken Ausbreitung in den letzten Wochen auf mehreren Kontinenten. Es geht nun in die "exponentielle Phase" (die Kurve an Neuerkrankungen steigt stark an).

Spätest im Herbst, werden auch einige Arivaner daran erkranken, mal sehen wer der erste hier sein wird. Erkrankung dürfte aber nicht stärker sein als eine "normale Grippe" sein, aber die ZAHL der Erkrankten und Toten dürfte deutlich ausfallen, weil es - im Unterschied zur "normalen Grippe" (mit der doch ein gewisser Anteil der Bevölkerung jedes Jahr Kontakt hat) - in der Bevölkerung keine nenneswerte Immunität besteht. Eine Belastung für die Gesundheitsbudgets dürfte sich in jedem Fall daraus ergeben.

Man muss hoffen, dass der (unwahrscheinliche aber mögliche) worst case ausbleibt und das Virus sich mit anderen Influenzaviren (zB den immer noch in Vogelpopulationen verbreiteten tödlichen Vogelgrippeviren) zu einem tödlicheren und zugleich kontagiösen Virus verändert. Wahrscheinlicher ist, dass sich das Virus in seinem globalen Seuchenzug über die nächsten 1-2 Jahre abschwächt und dann (fast) verschwindet bzw. irgendwann in späteren Jahren verändert wiederkommt.

der chart sieht zwar weiterhin sehr gut aus, aber mittlerweilen sollte es jeder geschnallt haben, dass die Pandemie unausweichlich ist.

Das ist mein erster Verfünffacher !!!!  Und das in so einer kurzen Zeit!

Bei Novavax gehe ich nun zu 3/4 raus (chart ähnlich wie Biocrystal). Roche (enttäuschend, aber ok) fliegt heute ganz aus dem Depot.

Ich kann zwar eine weitere Verdoppelung bei Novavax und Biocryst nicht ausschließen, aber diese beiden Aktien haben so hohen Depotanteil erreicht, dass mir ein ruhiger Schlaf wichtiger ist. Ich kann nicht länger zusehen..

mit ein bisschen Glück (und besserem Umfeld) gelingt vielleicht noch ein Sprung nach oben (trotz der minus 5 %) Obwohl ich gestern zur Hälfte raus bin, ist diese Position immer noch erheblich für mich, werde daher mit einem SL absichern - bei 5,90.

kleine Konsolidierung. Insgesamt ist diese Aktie nach wie vor stark! Neu einsteigen würde ich zwar nicht mehr, aber mit der verbliebenen Hälfte bleibe ich weiterdrinnen und ziehe das SL auf 8 (!) Euronen nach.

Wahnsinn, das hätte ich mir nicht erträumen lassen, obwohl ich immer davon überzeugt war (und noch immer bin), dass diesen Winter eine ganz heftige Grippewelle Europa und die USA erschüttert.

Bezüglich Biocryst: Biocryt produziert einen Wirkstoff, den man als "flüssiges Tamiflu" bezeichnen könnte, den man im Gegensatz zum Roche Tamiflu (tabletten) nicht schlucken kann, aber dafür intravenäs geben kann.

Fundamental habe ich damit insofern meine Probleme als man intravenös verabreichbares Tamiflu gar nicht (zumindest nicht im nennenswerten Umfang) brauchen wird. Daher auch meine Gewinnmitnahme in letzter Woche und mein völliger Ausstieg wird folgen, wenn das SL schlagend wird (oder nach einem weiteren stärkeren Anstieg). Wahnsinn fast 400 % seit wir hier posten!

Wart mal ab wenn die USA die Emergency Use Authorisation für die Krankenhäuser + Armee + Polizei für BCRX bekanntgegeben wird!
18 US Dollar nach der Bekanntgabe die nicht mehr lange auf sich warten lassen dürfte!

BioCryst loss widens, but H1N1 flu drug almost ready

BioCryst Pharmaceuticals Inc. reported a third quarter 2009 net loss of $10.6 million, as the company’s most promising experimental drug is taking critical steps to commercialization. The Birmingham, Ala.-based biotech firm’s net loss during the same period in 2008 was $9 million, according to U.S. Securities and Exchange Commission filings. BioCryst’s net loss through the first nine months of 2009 were $28.6 million. That is an improvement from its $34.8 million net loss during the same span in 2008. BioCryst employs about 40 in Durham, where the company has some clinical operations. While BioCryst remains in the red, it is inching closer to commercializing its first product. Its intravenous flu drug peramivir was recently given emergency use authorization approval by the U.S. Food and Drug Administration to treat certain hospitalized adults and pediatric patients with H1N1 flu. BioCryst (Nasdaq: BCRX) is completing production of approximately 130,000 courses of peramivir and is prepared to make more if requested by the U.S. or other governments. BioCryst has received 42 requests within the U.S. from physicians to supply peramivir through an emergency use investigational new drug process for eligible patients. In September, BioCryst was awarded a $77.2 million contract modification by the U.S. Department of Health and Human Services to complete development of peramivir.
